LC question
    #5424303 - 03/21/06 01:13 AM (17 years, 10 months ago)

I recently inoculated two jars of liquid culture made with powdered malt extract. I dissolved the powder completely to begin with, and used about 1 Tablespoon to 1 cup water. Now there is a grimey lookin sedimentation in the bottom. Anything to worry about?

Probably overcooked your LC. Make a new one and PC it for a little less time.

While Cappa is most likely correct concerning the impetus of your issue, I would contend (on the basis of the experience of SWIM) that it will be fine.

You used too much malt extract. A tablespoon is about 10 grams. I never use more than 10 grams per 4 cups of water and get some really nice, thick liquid cultures. I have found, in most cases, that the grimy sedimentation is mycelium feeding on the bottom. An excessive amount of malt extract can easily discolor your colonizing mycelium and give the false impression of a contaminate.

Next time I recommend using 10 grams of light malt extract and 10 grams of dextrose to every 4 cups of water. The results speak for themselves.

Right on. thanks for the feedback, i do think that I put in too much extract, and could possibly have overcooked it also. I recently checked the jars, and the sedimentation has turned into larger thicker more translucent clumps, so i think the mycelium is growing down there. Since it could give the false impersonation of a contam.... I could try it out on say a cake and if it grows its not contaminated right?
