Success making my first spore syringes

I wanted to report on the success I've had using a method I learned here that involves minimum attention to sterile technique. I use one of my 1/2 pint PF jars but seal the four holes with silicone sealer. Then I pressure cook the jar with the lid loose to sterilize it. 20 minutes at 15 psi seems to work fine. After the jar has cooled in the cooker, I screw the lid on tight.
When I get a specimen from which I want to collect spores, I cut off the cap and place it gill-side down in the jar using a pin or straightened paper clip to avoid touching the cap, opening it just long enough to place the cap inside. I leave the cap in 24 hours, then remove it using a wire to avoid touching the inside of the jar. I wear a surgical mask and gloves for this. I leave the cap loose with the jar turned upside down overnight to let the spores dry. Then I screw on the cap and store in a dark, cool place until ready to use.
I sterilized distilled water in 10 cc syringes in the pressure cooker, again 20 minutes at 15 psi. I kept the covered needle point facing upward, as recommended, to keep the water from boiling out. When these are cool, I clean the top of the PF jar with alcohol, and inject the sterile water forcefully onto the spore print and shake vigorously. When all 10 syringes have been injected, I then flame the needles, cooling them quickly in alcohol, and withdraw spore water back into the syringes, shaking the PF jar each time to redistribute spores.
I then injected the spore solution in pint jars of sterilized rye grain, shaking immediately and again after the spores germinated and colonize about 20% of the rye. I got contamination in two jars out of seven that may have occurred during inoculation since the other jars were clean. From one single cap of Penis Envy I was able to make 10 spore syringes. I could have made more if I had more syringes, but my pressure cooker only holds about ten at a time.

yea I am envious.... I am yet to make my own syringe... hopefully in the next month or so when I have a bunch of fruits..
and a PE at that!

I may have to use your tek to make my own, i also have yet to make my own. Today I made my first WBS jars and i am hopeing they will work. I have 8cc of B+ left that i am going to knock up 6 jars with and leave 2cc as a safty. I have a pressure cooker but no glove box. So you put 10 cc of water in and then PC with the needle up? Thanks again.

nothing personal...
but I hate the method of putting a mushroom cap into a jar and making a spore print.
I just hate scraping spores from the bottom of a jar.
its a pain in the ass!~

what method would you recoment to a guy with a PC and no glove box from after the mushroom unvails to spore syringe

To be honest, Roadkill, I didn't scrape the bottom of the jar. I merely shook it vigorously. The two syringes I've used so far both produced clean cultures, so apparently enough spores get dislodged this way. I can always scrape the bottom for the next batch from the same jar...
