if I sterizile a jar of wbs and use a synthetic filtration disc for a cap when I'm going to inoculate the wbs do I just sterilize the needle of a spore syringe and push it through the disc and inject my spores or LC? If so should I do this in a glove box? Do I need to wipe down the disc with alcohol?

a filter disc will retain hole shape from the needle if inoculated thru there, so that would defeat the purpose i would think. put a bead of silicon over a hole to create a healing injection site.

lightly mist your GB with 1:10 bleach water
then what I do is I have a wet rag or strong paper towel in there throw your jar and needle in also.... wait about 10 mins until the bleach water settles... and also throw in a cotton damped with rubbing alcohol
lightly wipe down the jar with the rag... make sure its not dripping wet!
then inject the spores right through the filter disc is what I do (wipe it down with cotton and rubbing alcohol) (if you think your GB is sterile enough then you can open the jar but I would not suggest it)
thats what I do when I use a GB.... you will prob get other answers from other peeps

will the silicone self heal? would it be best to prepare the filters with silicone injection spots even before I put them on the jars and sterilize?

Do NOT inject through a filter disk. If you use synthetic filter disks, you'll need to lift the filter up to squirt under it. Do this in a glovebox, or in front of a flow hood. RR

one other quick Q. How much headspace should I have between the wbs and the filter?
