Help On New Idea
    #5377223 - 03/08/06 11:05 AM (17 years, 10 months ago)

I made a LC using a new method of mine, and its working well (2 days in). BUT I don't really feel that using it to inoculate jars would be helpful. So I was wonder if I could sterilize some substrate. I was going to use brf/verm for now, and order some 50/50+ mix from the new place (mediummania). But do you think that if I put a 1 1/2 inch layer of substrate on a baking pan (heated to 400 then cooled), and then pour the liquid culture over the whole layer, and put a 1 inch layer of damp verm that it would grow easily, and fast?

I see MANY potential problems with this. The answer your going to get from most people is no.

Try it out, see what happens.

Learning experience?

Sure it's a waste of some time and a very tiny amount of resources but George Washington didn't invent the light bulb by sitting around and asking people if his experiments would work or not... (In fact I'm pretty sure he didn't invent the light bulb at all.)

Its so SO easy to make LC's, that it's not really a waste.

it's a waste when a perfectly good LC is put into use via a project that's doomed to fail.

I am all about experimenting with new things, but you need to approach new ideas a bit more scientifically/logically. You need to atleast EXPECT something to have a chance, and there is no expectation of succeeding here.

Try it, make a thread about it, and then when someone else comes along with the same idea, they can see specifically how it will not work. That way it isnt a total loss atleast, as it may help out someone that wants to try it in the future.

Ok here is an idea,

What if you took two trays that are suitable for the oven. Filled the bottom tray up with a substrate of let's say, a mixture of grains, horse poo, and maybe some coir to help speed up colonization.

Once the substrate is at proper moistrue content, place the second tray on top of the first one and sealed the two together with micropore tape.

Then you weighed the whole the thing.

Maybe poke a few holes in the top tray and cover those with micropore tape. Then place into the oven at 250 degrees ferinheight for lets say two hours.

After that time is up you remove it from the oven allow it to cool and weigh it once again. Find the differnce in weight and this should be how much water was lost during "sterilization". ( 1g = 1mL of water)

Once you calcuate the volume of water that was lost during "sterlization" add that same volume of LC using a LARGE syringe (availabe at farm supply stores)

Biggest problem I see with this is the substrate burning.

you cant sterilize grains in the oven, introducing that much LC to an unsterilized substrate is just asking for contams. Having uncolonized grains exposed to the elements is just asking for contams, using liquid culture on unsterilized substrate will just lead to contams.

See where im going with this? You're not creating a sterile environment, your substrate will be exposed to open air, and you will have uncolonized grains littered throughout the substrate. All of those factors, on their own or combined, will lead you down the path to failure.

If anything you need to make up some spawn bags and go that route, even then to be truly successful YOU WILL NEED A PC.
