So my Karo LC that I have been nursing for a week or so has nice little fluffy balls floating around in it. At this time, I have a poly-fill stuffed vent hole and have been careful not to shake the jar (to keep the poly-fill dry.) Instead, I just swirl it and get the marbles rolling around the bottom. Well the little fluff balls are kind of just sitting there. There appears to be no more of them being created. I'd like to know if I am supposed to be violently shaking this jar to break up the little fluffy gigs to get them to multiply. I feel that if I let this sit, it's not going to do much more.

I have the baby bottles going now, and can give them a violent shake without wetting poly-fill. I noticed that these containers have a copious amount of air bubbles on top and mixed in. There is no mycelium formation right now, because they are too young.

God I ramble on....anyway. Am I supposed to be shaking these to tear up the little fluff balls? I added a couple of pictures for reference.

Shake it but not that much, maybe once every two weeks from now on. Myc will grow anyway. If what you need is to inoculate, i think u can do it already but less myc will result in the LC, again, it will grow anyway. I always have more LC than i need just be patient u will have plenty of myc.

Don?t over shake it cause it can weaken myc and get contams :S

Shake it EVERY DAY. This oxygenates(sp?) the solution so that the myc can "breathe". If you look at posts by BlueHelix and Agar, they actually use magnetic stir bars and plates to keep the solution constantly shaking/stirring, which speeds growth tremendously.

OK...so can I take the jar in to the clean room and replace the poly-fill with micropore tape then? Then I can put my finger over the hole and shake like hell, then replace the tape with fresh, dry stuff. Does that sound about right?

i dont know about all that filter replacement... maybe if you have a glove box...

id say just giving it a gentle shake will suffice and a slightly damp poly-fil wont hurt too much if it happens, use double layered coffee filters over the top too.. I know the wet poly-fil might send ya some contam risks... but if you jar is only 1/4 or 1/3 of the way full its alot easier to not worry, and ya still got plently of LC.

Best to dip a piece of micropore tape in alc & set aside, remove polyfil in a G/B. Place sanitized gloved finger over the hole, shake shake shake:hairmetal:, then cover the hole with the alc soaked micropore tape.

The adhesive will not be affected by the alcohol? If not, that is very interesting.

Wet it will slide on, dry in a minute or 2 & stick like masking tape.

can't you just drop in a little h2o2 in the LC and not worry about contams during your filter change? in hte past ive used it like this and my place is rather dirty/ prone to contams. h2o2 kills spores but not myc. ive just used the stuff whenever i had to fool around with opening an LC. worked fine.

H2o2 is short acting and turns into oxygen and water very quickly, in other words more trouble than it's worth IMHO. FYI I prefer the micropore rolls over the tape dispenser as that leaves a section of the tape exposed to air (contams) unless stored in a ziploc, BTW I store most everything in ziplocs when not using. :wink: You could just pop the roll off the dispenser easily enough.

So I shook my LC and let her roll. Here is what the result was. I think I now understand why Agar talked about a "matt" forming on top. Apparently when an LC is properly aspirated, the mycelium is coated with air bubbles and will float. Here is what the same LC looks like after being shaken. In these pictures, I had agitated the jar to take the shot, otherwise all this mycelium was floating on top.
