cloning old, dry, frozen shrooms?

Hi Folks, Great site, unbelievable amount of information. I have spent many hours already exploring as a guest but have been unable to find an answer to my question, so I have registered and here goes... I have some dried mushrooms which are at least 1 1/2 years old and which have been stored in the freezer during much of that time. They are still potent and have the most desirable effects and only a few remain. I would very much like to save this genetic make-up. What are the chances of a viable clone taken from such material? My understanding is that it should be possible, fungal genome banks store genetic information in this fashion (whole dried, frozen fruiting bodies), or at least I think I read that somewhere, but now can no longer find that information. My primary concerns are that while the material has been frozen much of the time it has also gone on several long road trips. Also the mushrooms are physically quite small, most are broken, and of coarse they are very hard and crumbly, it will be difficult to obtain an interior sample. So I know it won't be easy and may take many passes on agar plates to clean it up, but is it possible?

Possible, but a long tough row to hoe.
A few spore syringes from a vendor(here)would be quicker & easy way to get started.

he wants to save the genetics though.

I doubt he will/can clone anything from that.
He may get some spore laden gill material to germinate on agar. Long tough row to hoe.
Only way to tell is to break out the perti's & go for it.

I'm not sure but I think the tissue dies when it is dried, and therefore will not grow. Something I have wondered myself, but just assumed it wouldn't work.

Here ya go. http://www.shroomery.org/forums/showflat.php/Cat/0/Number/2390329/page/0/fpart/1/vc/1 It's a long thread but worthy of a good read. Dry tissue can be saved, but I suggest hanging onto those old mushrooms until you get some experience growing and especially with the petri dish/agar work which is required. RR

I cloned a turkey tail mushroom I collected and dried. Powdered in a coffee grinder, soaked a few minutes in 1:10 dilution of 3% h2o2, put tiny pieces in wide mouth pint of brown rice that had 10 mls of undiluted 3% h2o2 mixed in. All the jars (10)contaminated but one. The last one became contaminated but the turkey tail overran the contaminant.
If I had sterile conditions I'm sure petri dishes and frequent transfers would work better.
Turkey tail is a polypore and they are know to be tolerant of drying out and getting rehydrated while growing.
