Bunk Spores???

Hey guys got a question. I am on my second attempt to grow with BRF cakes. I ordered my sryinge from spores101 and well i have had no growth. The new jars have been inoc'ed about 3 days ago. I know that that is not longth enough to start making assumptions but, I also have a LC (honeywater) that i knocked up about a week ago. I see growth in it but hardly any at all. So whats the deal here. I followed instructions to the T. Am i just doing something wrong. Its getting me frustrated. Thanks for any help you can offer. OH BTW my strain is Mexi-Cube.

What is your LC in, a baby bottle, a mason jar? How are you providing gas exchange to your LC?

My LC is in a Ball Mason Jar. I has layer of Tyvek then a polyfil filter hole in it. i got told that provides anough gas exchange. and its just one hole. about 1/8 inch in diameter.

ive had a bad experiance with this company, not their cubie spores thogh those worked fine.

So should i just order some more spores. I was thinking if i did to go through ralphsters this time. I read good things about him on this site.

In my experience the hole needs to be a minimum of 1/2" to be of any use. I don't think you need Tyvek and poly-fill, seems a bit redundant to me. Make sure you're agitating the LC everyday for optimal results. If you have the means, I would make an effort to increase the size of the hole and agitate everyday.
Most liquid culture failures can be attributed to improper gas exchange and lack of agitation. The other substrate you inoculated sounds normal to me. Spore germination can easily take 3-5 days and is not unusual to take 7-10. Use a flashlight to look for mycelium in your LC. Sometimes they're there, just harder to see without proper lighting.

Alright i will make a bigger hole. Would it be ok to take the lid off. And should i use tyvek still and just eliminate the polyfil or neither.

You can take the lid off in a sterile glove box. NEVER take the lid off any substrate in open air. You'll catch a contaminate quicker than you can spit.
Using Tyvek or poly-fill will suffice. I personally prefer poly-fill but its up to you.

My first question would be what are your incubating temps at? Because they should be in the low to mid 80's also don't forget to shake the syringes although a couple of spore sites use ultrasonics and a suspension agent so shaking is no longer necessary. GL

I got a small TiT holds 6 pints jars. (Wanted to start off small) and yes i keep it at about 83 in there. and another yes, i did shake the syringe. I hope this works because i dont want to buy another syringe. I want to make my own. Do my BRF jars need to have a big hole in them also for gas exchange?

The holes for your inoculation points will provide enough gas exchange. You should have 4-5 in there, which together will provide what you need.

even if the hole is small like 1/8 inch in diameter. Also why couldn't you just leave the inner lid off and use a tyvek lid. then seal the inoc holes up with micropore or something, or would that provide to much exchange and dry out the cakes.

If you have multiple inoculation holes they will equal the appropriate amount of air flow needed when combined. You don't want to just have a Tyvek lid for an opening. You'll experience excessive evaporation of moisture and dry your substrate out. The inner lid is required.

I had the same problem with Spores101. Bunk spores. Everything went smooth, but not a single bit of growth, not even bacterial.
