What would happen if you placed a piece of cloned material on one end of an agar plate, and spores on the other and allowed it to colonize?

Each sub-strain would compete for nutrients. My guess is that the clone would have the advantage. It wouldn't have any competition on its side of the plate; thus would grow faster and dominate the plate. Then I think you would see a line between the clone and multi-spore mycelium.
I've often wondered if the mycelium of the same species would cannibalize the weaker of the two, taking over the substrate.
And, I think you saw this coming, try it and see. Then post results.

i think the tissue would create mycellium faster then the MS innoc. Ive seen agar plates that had two strains on them, there was a clear divide in the middle of the plate between the two strains.

Nice pic ohm. That shows how you can see the two identical cultures growing together. The one I posted above is of different strains, therefore you can see a line of bare agar between them. RR

you know what i just thought, basically the line cant really be of bare agar.
reason i think that is, if it were of bare agar the two culture would not know of each others existance and therefor continue growing - but as they know, they dont.
therefor there must actually be some mycelium of both cultures in that "bare" strip, despite being invisible or hardly visible. peace ohm

See http://www.mushroomvideos.com/868246.html and look at video 3. Look how far away the mycelium is from the contaminant when it begins to 'circle the wagons'. There was no mycelium in the no mans land, so I have no idea how the reishi knew of the presence of the contaminant-I can only speculate about harmones, scents or actual sentience. However, you are correct. In the picture I posted above, there are small, invisible to the naked eye, mycelial strands that reach out ahead of the growth(I call them 'scouts') and when those mingle, they stop the respective growths as a survival tactic. It's these 'scouts' that are harvested once they make contact for hybridization experiments, when one is attempting to breed dikaryons. I think it's interesting how the more we learn, the more we realize how little we still actually know about our beloved fungi. RR

Video number 5 (reishi fruiting from substrate bags) was really cool. Interesting how the initial growth out of the bag almost pulsates, growing and resting alternately.
