Colonizing from Agar

I have some concerns about colonizing my rye with agar.
Im jumping up to the rye substrate method;
agar is almost colonized 100% (PDA agar)
What sort of method would you use for gas exchange in the 1qt mason jars? would i invert the rubber seal lids and screw them down? leave the rubber down and loosely screw?
my incubation chamber is maintained at 27 degrees celcius (which is about 80 degrees F i think)

Use either a filter disc, a tyvek filter or a polyfill filter.
You?ll find instructions on how to make a polyfill filter in the link below.

AGAR AGAR can be purchased from healthfood stores, natural food stores, etc...
they have it in jars next to jars of other powdered, or dry herbs and spices.
Agar can be purchased from several vendors here, with the Malt already mixed with it MEA.
Agar can also be purchased from biological supply catalogs, but it tends to be expensive.
Cheapest route is to make your own, Agar Agar and tan Malt.

I make agar from boiling about 4 good size potatoes chunked up real well for like 40 mins, then strain the water off them (SAVE THIS) disgard the potatoes..
then throw in ~15 grams of dextrose powder, 2g of yeast, and i use malt extract too (~5-10g) and 15-20g of agar agar. i paid like $4 for 50g of agar agar.
boil this stuff up, and either pour into autoclavable dishes, and a/c,
OR a/c the whole thing for like 40 mins, and then pour it out into sterile plastic pitres
ive done both, both work well.
agar can be stored after a/c'd.
Oh, i forgot to mention, after boiling the water, either use 1L of it, or if you dont have enough, add water to bring it up to 1L

sounds good, that's actually the mixture i was going to use anyway. Are you using this technique for mycellial culture and then extraction, or spawn?

Oh lord no. This is merely for spawn, and for increasing cultures.
also, i am going to try the liquid mycelium by mixing 4g of malt extract with 96mL of water..
and then inoculate this with some mycelium
what do you think?
has anyone ever used this method (liquid cultures?)
