help!! major contamination problems

i have been growing cubensis for three years and just recently, i have had big, big problems with green mold. First i clean and wipe down the counter and around the room. I then use a mixture of 4 cups coarse ground verm and 1 cup brown rice flour and 1 cup water. I use a pressure cooker and cook the jars at 15 psi for 40 min. I then sterilize the spore syringe needle with a lighter and inject cakes with spores. In between cakes, i use a cotton swab with rubbing alcohol to clean the needle. no matter what i do, mold will grow top to bottom, and occasionally where the needle injected spores. I have got brand new syringes and sterilized them and STILL have problems. after making something like 50 cakes, only a pathetic 5 are lucky enough to survive. I am just about ready to throw in the towel becuase no matter what i've done, nothing seems to work. . . HELP!!!!

p.s. I also have problems with my recepie for substrate causing the mixture to become a little mucky and slow down myceluim growth.

well I can give you two more things to help cut down contams, the oven tek and lysol...other than that your procedure is sound from what i can see.

Also..who is your spore supplier? If they are all from the same source then it might not be your fault at all.

Maybe this will help, Im sure you know most of the teks and you probably know everything i will say, but Maybe you can use it as a double check list.

1. Some teks suggest pulling air into the syringe so spores can be shaken. But pulling room temperature air can and probably does contain other bacteria and other spores. Which will contaminate the entire syringe

2. Try using polytek, or another filter of some sort.

3. Keep the jars in a sterile environment if possible

4. Keep the substrate from being too watery. If you can squeeze water from it, it's probably too wet. wet substrate attracts molds

5. use a top 1/2 inch of dry verm ontop of substrate to further protect against contams

Make sure that you are letting the jars slowly cool while still sealed, tightly, in the pressure cooker. I usually just turn off the heat and walk away. A good 3 hours later and i'll take off the pressure cooker lid and tighted the jar lids, then close them in the pressure cooker again, and then innoculate them 2 or 3 hours later. I've also had success with waiting untill the jars are fully cooled down, taking them out, then tightening them and then innoculating.

This may be overkill but here's a few ideas:

Before you mix your substrate:
Presteralize your substrate 20min/300deg in a
corningware/pyrex bowl w/ lid.
Do the same with your verm about twice as
long, you can throw in a couple of spoons,
knives, pie server ect with metal handels to
work with.
PC your jars 20 min and Boil your water.
Do this where it all comes out at about the
same time. Take the stuff out of the oven
and set them outside(covered) In 30min to
an hour it will all be cool enough to handle
(given it's cold outside). Wash your hands
with anti-bactirial soap and go for it.
Put your flour and verm in new ziplocks and
use them as you need them.
Before you innoculate give your jars/tape a
quick rubdown with alchohol.
When you receive a shippment of goodies from
your supplier. Resist the urge to open it right away.
It was probably packed in a semi-sterile enviorment
but no telling what's on the outside of it. Wash your
hands good, handle the package as little as possible
to get it open, and place your syringes/prints ect itno
a big ziplock until you are ready to use them. Then
you can look at them all you want.
During the (somewhere around) two week process
of watching cakes colonize, there is only one or two
days that you really effect what's going on inside.

Like I said it may be overkill, but, I have an eighty lb.
pit bull in the house and haven't lost a jar to contams
yet. Hope that helps! Good luck!

Work on the substrate, it should not be muddy. Each time I prepared a muddy substrate, i had troubles.
