Trouble w/ S. Africa Transkei (broken glass in water jar)

okay, so, usually I use colonized Thailand Koh Samui Super Strain on WBS, take a chunk of it with a spoon, put it in a half pint w/ broken glass and water, shake to knock mycelium into the water, suck up syringes and use this to inoculate my wbs jars. This has always worked for me. Now, i used south africa transkei for my last batch, wbs prepared the same as always, and I have no growth at all after a week. advice? maybe that method works with some strains and not others

Why don't you just do a straight grain to grain transfer and skip the syringe?

temps at 80. I havent tried grain to grain yet b/c I havent built a glovebox yet.

You don't need a GB for GTG. Be extra sterile. Oust and bleach EVERYTHING. Sterilize your utensils you'll be using between each transfer. Be as quick as possible. I just transferred 4 pints of colonized WBS to 20 quarts with no contams and no GB. A small closet works great for a work area.

being extra sterile requires a flow hood or glovebox, open air transfers have a risk, oust, bleach and what ever dont actualy sanitize the air, and as cheap as it is to make a box for transfers, it's pointless not to.

I guess you're right. I suppose I just got very lucky the past 3 times I have done this with 10-15 quart jars per run with no contams and no flowhood/glovebox. I'd love to have a flowhood, but the girlfriend's not to keen on turning our kitchen into a lab. Whatever works for you man.

luck may play a small part but good technique is also a major factor, people have been known to get contams even with flowhoods because they have no real concern about the processes...

yeah, i want to make a glovebox and do grain to grains, but thats out of the question for a couple of weeks. Ive always had success before with the broken glass in a jar, so i was just trying to repeat that until i build a gb. im still trying to see if anybody has had success using this technique for some strains and not others
