WBS Agar?
    #5256050 - 02/02/06 04:29 PM (18 years, 11 months ago)

Would it be possible to use WBS as the "base" for agar? I'm really wanting to start isolating a sub-strain, and I have read that mycellium becomes selective over time to the type of agar used. Since I always use WBS, whether to spawn or case, I want to try to make plates that are as close to my substrate's nutes as possible. Could I substitute WBS for potatoes in PDA. Maybe simmer/steep the seed in water, drain, add my agar powder and a little honey/karo? Anyone have experience with this? I've heard of people using dog food to make agar, so why not WBS?

Perhaps WBS Flower. (Grinded WBS) But I doubt it would be easy to work with. Maybe make some wbs flower, strain some water through it, then sterilize the WBS Flower enriched water, and then use this to hydrate your agar powder.

Hey Frag. That's an idea. I hadn't considered just adding ground WBS. I'm going to do some experimenting. Maybe try simmering some, and try powdering some and see which way works the best. Anybody else messed with agar base alternatives, in particular, WBS?

There's a tek somewhere on this site detailing how to make a ghetto (agar-ish) base with just mixing brf with water and pc'ing. I assume you can do the same with wbs flour.

My question is: why do you want your mycelium to be selective? If you keep breeding myc on the same nutrient base it won't have the enzymes to break down other types of nutrient bases, it doesn't necessarily mean it'll break down the same base BETTER.....Or am I misinterpreting? I'm sure transfering myc from PDA to sheep's blood to LC to grain to poo to w/e will yield a mycelium waaaaaay more adaptable to nutrient environments than wbs to wbs to wbs.
