Possible LC contamination

First of all let me tell you that I'm pretty good at spotting contaminates in liquid cultures. This is kind of throwing me for a loop though. I have a few liquid cultures that are showing signs of what I construe as mycelium, but appears to have a tissue structure.

The areas of concern are white, round, and floating at the top of the LC. The appear to have some characteristics of mycelium, but I'm just not sure. These liquid cultures were made from light malt extract and dextrose. This particular one has been incubating for 16 days.

Here are some photos. I do apologize, as some of these are not quite as clear as I would have liked.






Any insight would be greatly appreciated. Thank you.

P.S. This is probably the longest I have ever incubated an LC. Maybe these guys are just overdone. Just a thought.

I have had 1 or two jars do the same. I simply left it to see what happened. One turend a nice dark green color, the other one just remained white. So i am guessing it could be mycelium. I would definitely test out that jar. On some grains first. I am going to do so.

Thanks for the first hand knowledge. I had two other jars that are also looking like this and one of them is turning green on me. Damn it! Oh well. The nice thing is that the contaminate, if that's what it is, appears to only be localized to the surface. This may provide an opportunity to extract mycelium without the contaminate. Kind of along the same lines as using agar to clean up a print.

I have had a lot of these recently. I would not chance trying to get good myc, since it is a liquid it is far easier for it to disperse throughout the liquid. I would just toss it.
I am no longer using jars with filter discs, they nearly all fail on me lately.

16 days does seem massively long, isnt it only supposed to be for the most a week?

I have had LCs take a month to show any growth. usually it is 1 week after the first signs of growth.

That solution is too dark for a LC. 16 days incubating is also too long. If in doubt, throw it out, I would. You wouldn't want to waste another month knocking up jars and waiting for myc to show up.

If it's snow WHITE. It's right. :grin: :thumbup:

If you don't stir or agitate one every day, myc gets to the top & forms little islands that turn to thick pancakes. As, it has better gas exchange on the surface.

How, I test for LC contams, is to STOP stirring one.

If anything grows on the surface that isn't snow white.

It isn't RIGHT.

Most often contams show up as light gray / light blue growing on the surface. :mad2:

Pink on an LC surface is most often either Cinnamon or Lipstick mold :mad2:

Thanks Agar. I'm waiting on stir bars right now and I've been neglecting to swirl them around. I had a few pure white areas that have turned green on me, but that one seems to be doing just fine. I appreciate the photos.

16 days is a bit long but I've let some liquid cultures go for this long before and they have serious concentrations of mycelium. They make for really good growth in spawn bags.

Gema, as far as the color is concerned, the color is correct. I've made probably 20-30 liquid cultures using light malt extract and dextrose and they all have this consistent color. My typical incubation period is 7 days. I use 10 grams of each ingredient per 4 cups of water. The color is likely derived from the consistency of the light malt extract.
