Printing to LC is tricky, and although I use LC almost exclusively I wouldn't suggest anything but agar for prints. Not that it can't work but there are more vectors for contams. Using an inoculation port for multi-spore, or a glovebox for clones (into an LC with inoculation port) is extremely safe in my experience provided the procedure is sound.
This tek is 7 years old... anyone who uses it, I would recommend investing in a bag of polyfill and tube of silicone. A piece of automotive vacuum tubing can also be useful for getting the filter up off the lid.

If it didn't work this thread wouldn't be 30 pages long but time of year and location determine what's in the air so anything is possible including contams. Removing contam vectors seems like a good idea to me and the DIY route is cheap and easy.

I didn't say not to use LCs, just suggesting an inoculation port and filter.

Conditions are not correct for germination of spores in sugar LCs
My spores seem to like honey a lot.

Also sugar based LCs are not the proper environment for spores to germinate
I pointed out earlier in this thread that this comment was odd. Spores germinate fine in various sugar solutions. You might want to provide some rational info to back up your claim, otherwise you just seem biased. In this case 'proper' seems more like a (wrong) opinion on your part.

All that doesn't prevent the spores from germinating and consuming the sugar.
I do several batches of (honey) LC per year and haven't had a failure in 5+ years. Spore source, procedure, and medium container all play a part in a person's success rate. Not all situations are proper for LC but that's different than saying sugar isn't a proper food. I personally don't care if everyone forgets LCs exist. I have no reason to promote specific mycological practices, though I can understand in light of a failure rate having a preference and promoting it based on the desire to see others do well.
But my experiences differ, so I say sugar water is a good medium for colonization from syringe which is what most people use it for.

mycofever said: In my trials I have found that if the spores do not germinate than you have either mixed your lc to rich or sterilized for to long. Sterilizing for too long will caramelize the sugar I have done this a few times and had zero growth.
Same here on the few fails I've had. What are your PC time/temps?
1.3 Tablespoons liquid sugar per pint of water will make a 4% sugar solution. A 'heaping tablespoon' is close enough. I like to start with hot tap water so the honey dissolves quickly. I've always done well using sponsor syringes. I also use LC to clone tissue. Never a contam. If contams were a problem I would surely have moved on by now. I do advocate buying/making inoculation ports and gas exchange ports as it's easy and only costs a few bucks.

Good to know you can get a clear cook at 15 PSI. I used to run @ 10 PSI for 15 minutes, no problems but bumped it up to 20 minutes because it's closer to the holy grail of 250F for 20 minutes. The cook does seem a little more amber than the 15 minute cook, but it colonizes at the same speed. I'll try your method next time and see how it looks.

The LC should be thick by 2 weeks. Probably a bacterial contamination unless you weren't agitating (oxygenating) the liquid. I would assume a contam and start again.

Ahh yes.....gotta love that LC......besides being a complete waste of time, what is the point?
It's an easy way to culture multispore. I've been using it for years -w- a silicone injection port and polyfill stuffed vacuum tubing for air exchange. Works great.

LC has always worked for my purposes. I've used the same Shroomery sponsor for about 8 years now. My jars have an RTV inoculation port and poly-filter elevated with some gasket tubing. PC @ 10 PSI for 20 minutes. Syringe in open air and clone in a glove box.
Having said that I should mention that I have other hobbies that I do push technically. With mushrooms I don't care to mess with agar or advance the hobby further in any other way. Everything has been thought through to require the least effort at every step. I get consistent results, lots of shrooms, happy.
