The Rookie Begins! A question about substrate.
    #517867 - 01/11/02 09:13 PM (22 years, 3 months ago)

Today my friend and I started down the road to mushroom cultivation. It's kinda scary, not knowing if it's gonna work and whatnot. Anyway, today we made the substrate and filled the jars, but then realized that we didn't have anything to clean the kitchen with. Will the water concentration be affected by letting the jars sit overnight? We didn't have any time to sterilize and inoculate them, so we taped the holes and went out to get some alcohol and lysol. Would it be OK to inject about 2-4cc's of water in each jar before sterilization to wet them some more? We're using 1/2 pint jars and the PF-Tek. Thanks for whatever input you've got!

do whatever the hell ya want to them before sterilization...

Being extremely clean especially with brf jars is not extremely important until inocculation.

Seriously the pf style tek is sooooooo easy to do... you really have to fuck up bad to mess up..

Out of 20 jars I think I got 2 contams...
Mind you my house is very dirty and dusty

Just some advice though, do not put too much water in the jars, too much water is what usually causes contams. Just follow the instructions on the PF TEK for how much water and all. Yes use the over tek, its the simplest and best way to insure contam free air.

dude,

You better read what you posted again.
You do not inject the spores before you sterilize the jars.
Don't put more water into the jars than the tek calls for.
Tell you what....follow me
If you have already made up the jars....sterilize them now.
They need to cool down over night anyway before you inject the spores.
What are you going to do with the substrate boil or pressure cook it ?
Either way you are gonna wanna let it cool down.
If you put your spores into a hot or warm jar....your gonna kill the spores.
So make the substrate tonight....let it cool down.
Clean the kitchen.....bleach works good too.
And inject the jars in the morning after they have had time to cool down.
Read the PF-tek very closely....might even be smart to print it out and follow it.
Maybe you can work on setting up your incubator while you are waiting.
If you need help.....yell

Well I'm not stupid. I've read the PF-Tek more than a few times. I'm just a little worried that the substrate may get a bit dry. Anyway, it's too late to sterilize them now. Anyway, does it really take all night for the jars to cool? I'll be steaming them not pressure cookin', wouldn't it only take like 2-3 hours fot the jars to cool?

listen to Roadkill DrD, if you want to wait, 2-3 hours after sterilizing your jar to innoculate, have at it, unwise i think, but whatever floats your boat. 8 hours or overnight is much safer when it comes to preserving the life of your spores. what im sensing is impatients, this is not a rush project, if you rush and cut corners, your just setting yourself up for failure. if you follow the pf tek, you can't dry out your substrate unless you seriously cooked your jars, but then experience has always been the best teacher. good luck.

I recieved my syringes on Dec. 11 and harvested my first 50 wet grams last night , Jan. 10th. I followed the PF tek using a rubbermaid with perlite. It was surprisingly easy.

Depends on the tempeerature and the air movement in your kitchen.
2 hours is definitely too short, sonce you should leave the jars in the port while cooling to minimize the risk of airborn contamination.

When you prepare the substrate, don?t let it sit more than a few hours, since the growth of molds and bacteria begins the moment you mix the substrate.

If you leave the jars for more than a day or two unsterilized, they will most likely turn green.

And sitting them for a few hours before pressure cooking WON`T alter their water content.

Thanks a lot guys. Some of my nervousness is gone. Anyway, owr jars were done with sterilization at exactly 1 pm mst today. And we inoculated them at appoximatly 6:30 pm mst. Now I'm just sort of worried about contamination, but I really dont think I NEED to be. We were pretty sterile (used oven tek and sprayed EVERYTHING with alcohol), so hopefully, everything will go good. WISH ME LUCK!
