Jar conditions for colonizing
    #514025 - 01/08/02 02:07 AM (21 years, 8 months ago)

It's been two weeks since inoculation and nothing has happened inside the jars. Not contamination, not mycelium, nada. Today 4 of the 12 jars were opened up and checked for any internal growth, and there was none. The cakes seemed very dry, so water was added to the surface of two of them, and the other two were stirred and had water added. They were so dry it was hard to break apart the clumps. The other 8 were left alone in case these 4 go green in the next couple days from doing this. Any suggestions for salvaging these jars? Thanks

Yeah.... so just take the easy way out. Is the dryness the most likely cause? Maybe jars were boiled too much (1 hr). Not sure what happened. Thanks man

I previously boiled my jars for an hour with foil over the top. I never had problems w/ moisture levels as long as my moisture was correct to start with.

Are you using a dry heat source for incubation?

Humidity for incubation was approximately 40-60%, just in a sterelite container. Foil was also used over the lids when boiling. Substrate was prepared according to the directions, 3 cups of water for 12 half-pint jars. Not sure what could have happened. Perhaps the conditions were sterile enough, but the spore syringes (from sporechicks) were bunk.

Thanks for the help.

did you wait long enough to allow the jars to cool? if you didn't you killed the spores when you innoculated.

peace, ar393

Also, if you released the steam from your PC before
it completely cooled down, it could have steamed
the moisture right out of your jars.

This is probably what happened. The pot was opened to check to see if there was splashing a few times during the hour. The gasket to the edge of the lid for the PC was warped when bought, so it had to be used as a normal pot.

The jars were allowed to cool overnight - but the directions indicated that during inoculation to work on an open stove set between Warm and 200. It seemed hot after awhile and the purpose of this procedure was not clear (probably another sterilization tech). The heat was turned down, it didn't seem right.

Thanks for help.
