H2O2 CLONE TEK
    #512316 - 01/06/02 05:31 PM (22 years, 3 months ago)

Take your favorite mushroom and cut off the cap and the bottom inch of the stem. Put in blender with 4 cups of water. Blend for 10 seconds. Add one cup of H2O2. Blend for another 10 seconds. Innoculate 4 dozen quart jars with the oven tek by just pouring some blue slurry into each jar. Shake each jar for a minute. Within 5 days they will be fully colonized.
1.clean blender glass, top and bottom with a paper towel and isopropel alcohol.
2.When making the grain keep them on the dry side. You will be innoculating them with over 10 cc of blue water.

This is by far the easiest method of clone spawn production I have ever tried.

any questions? PM me

nest time i get a good specimen from my neighbor's cat i'll try it. my attempts at cloning have all resulted in jars full of mold, because i used honey water, and there is too much exposure to contams in my culture area.

blending for several 2 second bursts is better than 1 10 second burst. Your using too much peroxide, less would speed things up even more. For more sterility, don't use a blender, make a homemade eberbach container, use a quart jar and a set of blender blades under a jar ring. You can pressure sterilize it, unlike a blender pitcher.

I just looked at my profile and realized I had a website at one point in time on geocities, it's not there anymore and I have no idea what I had on it. Anybody remember my website from several years aga? PM if so please.

Would help reduce contams if ya washed the mushrooms before blending also. The H2O2 will take care of the rest.

Peace

Theres a "detergent" used specifcally for washing vegetables that I'm sure would work for mushrooms. Its advertised through TGIFridays commercials, i forget its exact name.

Washing/rinsing is more or less a precaution, not really necessary. Would just help eliminate all the contamination on the "skin" of the mushroom that has collected while growing.

Peace

Arent u supposed to use the bottom of the stem? Arent u aslo supposed to cut the outside of the stem away so any contams on the outside are not brought into ur myc. water?

You are correct but that is a different technique (the commonly used one).

He was introducing a new technique. He avoids the contams by mixing H2O2 into the mix which eliminates contamination but allows the mycelium to survive.

Hope it makes more sense to ya Man.

I think That mycofile's Idea is good in theory but neglects one problem. When the ederbach container (Jar with the blender blades inside) is sealed and blended while containing H2O2 Oxygen gas is realeased causing pressure to increase inside the jar and possilby explode. Anyone who doesn't want to ruin their kitchen, hand, and at the very least their shrooms should not attempt to blend any container that is sealed and contains hydrogen peroxide. At the very least they should be aware of this problem and continually vent the jar to releave some pressure. I can not stress how important continual relief of the pressure is even after many minutes of blending there will still be a pressure build up that could ruin the whole adventure. Be very careful if you are going to use a sealed container to blend H2O2.

I think trial blazer means wash (dip) in h2o2 .
and thats the main thing i would change with this tek is two add the h2o2 first thus washing the tissue. also i would not use so mush liquid, unless you had alot of tissue. mybe like a 4th cup of h202 at most and a cup of water at most for one stem.

Actually, this isn't my idea, some other dude started this thread. But you're right about the pressure.

Peel skin. Remove inner piece. Wash with peroxide (3% is fine here). Blend with sterile water in jar. Add peroxide.

BTW, peroxide doesn't protect a liquid culture the way it does a solid substrate. The peroxide is destroyed by the mycelium on contact. In a solid substrate, the peroxide that isn't in direct contact with myc remains. Don't depend on peroxide to last any length of time.

bumpage-foaf tried the no agar, no flowhood easy cloning method from the mycodirectory v 4.0 cloning section. has anybody else had success or failures using that tek. it has been 24 hrs and still no sign of growth. the h202 foamed like crazy when stem was blended. the blue puree was injected into millet jars. i know patience is a virtue but one of the selling points to this tek was rapid colonization. at this point it seems slower than honey h2o innoculations. does the h2o2 bath stunt mycelium growth?

manson or anybody- do you think it was the dilluted h2o2 instead of straight that made it work or the substrate in the little jar. what you described added up to about 10 days which is not that much faster than a multispore innoculation straight into millet qts. the flow hood sterility issues are what my foaf was trying to avoid while still having the advantages of a isolated strain. by the way 48 hrs and no sign of growth not even contams.
