Ss..ss..sss...six bills... My God why dont I just make a fucking NASA lab in my house.

Anyway I had this blovebox idea a few weeks ago, tell me if it will work. It seems the cheapest/easiest way to me. Im not sure what is the best way to filter the incoming air is, Iv'e seen dust masks, polyfill, and coffee filters with other designs but not all 3 of them at once. Suggestions welcome!

Well uploading that pic is being a pain in the ass and I cant properly convert it, but it simple so ill just use words:

Get a fan and a cylinder that the fan fits nicly in. Behind the fan in the cylinder put dust masks/coffee filters/polyfill to filter the air. Cut a hole the diameter of the cylinder in the bottom of a clear garbage bag. Attach the cylider to the bag (the new hole). Some clear garbage bags have a drawstring built in but if ur bag doesnt just make one (doesnt necessarily have to be a grabage bag either). Cut holes and attach long gloves in the side of the bag. Lay the cylinder on its side on a table, turn the fan on, open drawstring, insert working materials, sinch up drawstring, put hands in gloves and disenfect the inlfated bag. Wait a bit for the fumes to dissapate, then ur ready to go!

god damn. i thought that would be like $100-$150 at most. I was hoping for around$80. why the fuck is it so expensive its just a fukin plastic box with some gloves? what assholes. Everything these days is too god damn expensive. You know that shit costs them like $8.00 or less to make.

I estimate it costs less than $50 for materials for that box, minus tools. Probably takes them about two hours to build it. Talk abuut a rip off...

I made a glove box for free (had everything already) and it works great! Probably costs about $5 to make.

Items:

~Rubbermaid Container
~Plastic Grocery Bags
~Duct Tape
~Latex Gloves
~Saran Wrap
~A cutting device

1. Cut two holes on the side of the container near the bottom, big enough for your arms to get through.

2. Put grocery bags through the holes, seal with duct tape on outside to hold bags in place

3. Put holes for your hands in the bags on the inside of the box

4. Cover the open top with a layer of saran wrap, duct tape down to the side. (The wrap should not cover the entire hole..there should be a gap to put stuff through)

5. Sterilize Box

6. Put stuff in box, cover gap with another layer of saran wrap...overlapping the first one.

7. Put on latex gloves, put arms through plastic bags and work!

For those of you too lazy to make one, I will be selling this box for the low low price of $1,000 US. Very cheap, act now. Never again will you see a price this low! We cut the costs and pass the savings on to you!

You could use a dremel tool to cut the plexiglass in to shape, glue it together with silicone glue or aquarium glue or something like that, with hinges and weather striping on one side/top/back/what ever.

heres what i did.

i went to wal mart and got the smallest hepa quaility room air purifier.
the holmes unit cost about $45-60.

cut a hole in the back of the box that fits the output of the filter unit into the box and the intake outside.

I put a plexiglass lid on it, and cut two arm holes in the side.

when you turn on the hepa, with the output on high, clean uncontaminated air is shoved out of the arm holes and any other small holes in the box. with a positive pressure in the box, its nearly impossible for contaminates to be introduced. i just wash my arms and hands real well, put on a pair of big dishwashing gloves, and go for it.

granted, $50 is a bit to shell out, but i havent seen a contaminate yet.

When you install the heppa filter to the back do you install it toward the bottom of the box or top? The reason I think this is important is that I don't think you want a breeze across your work area and the heppa filter puts out a lot of air. If you install the heppa toward the top the air at the level you are working in would not have the air blowing over it. right??
I'm in the process of building a glove box from plywood with a plexiglass viewing panel. I plan to put several coats of paint on the inside and to caulk all the seems. Am I on the right track? I'm just not sure about the output of the heppa directly on my work area.

the output on mine is about halfway up the bacl of the box. the air is contaminate free assuming you havent contaminated it somehow or your filter is too old, its the same principal as a flowhood. but with a contained enviromant.

enter- theres no writeup in tmc or ggmm (2d edition) on how to constuct a glovebox, but there are a few pics. heres one to which i was referring in the Vendors/Gloveboxes thread you posted in vendors..
... see, the hard part is drilling the armholes and securing the plexi

the mushroom cultivator also has (in its appendix) a writeup on diy laminar flow hoods, which i wont scan and post but has been discussed in length at other times... with the proper equations and such for the blower, etc.

if you wanted, you could always try and make the glovebox yourself out of plexi (seal the edges with silicone or aquarium sealants)... but the hard part again would be the circular cuts in plexi for armholes... cutting plexi is tough. we have a local plexi/glass etc shop here, and they wouldnt do a job like that for us for under a certain amount (he woudnt say over the phone, but intimated it would be quite expensive and would take 2+ weeks... but they re also the only place around here that could ahve done it, so maybe thats why theyre expensive...)

you could make the whole thing out of plexi by sealin it together with silicone/aquarium sealants... except the armhole side, which could be ade of plywood (easier to cut and attach armhole thingys to) but you lose a bit of the field of vision within the box. but most work with them ona table, looking down, so the loss shouldnt be that bad. the plywood side could be attached with hinges, since thats the place where youd want to load stuff in and out of.

something premade? check out ebay and car repair places (that cater to car repairmen) for sand blasting boxes... theyre very similar to the sci supply design one, but of course not all see thru (but at 10% of the cost- they usu run about 80-100, and sometimes you can get free shipping). anchor supply hardware (has an online place too) has them, and often bundles a free ship/no sales tax thing. or a vendor could find a wholesaler to drop ship from- but then again, 100 is expensive for this hobby (comparitively) so i dont know if thered be interest in the prefabs

Nice! I love illustrated teks. Wish I had of seen that before I built mine I wouldnt have got the foil vent tubing. One question though isnt the wood lid succeptable to contams? It might be good to paint the wood with some kind of latex paint to aid in cleaning. But if you said it worked than it might just be overkill. A few usefull things I found out: 1) plexiglass + hole saw = melting plexi/stinky room 2) cutting holes by hand in plexi can be a bitch if you dont have an appropriate saw. 3) Vinyl gloves are the worst fucking things in the world for use in a glovebox. They tear very easily and are difficult to get on/off if your hands get a little sweaty. Latex gloves are 100% better.

it really only cost 50$$$ WOW now thats cheap well considering that thing was a piece of shit compared to my glovebox that i built for hmmmm bout 80$$$ wow only 30$$ more and it looks like that OMFG.
Wait till u see when i'm done, i still have to put the plexiglass and door on.

just use a 4" hole saw drill but to cut the holes plexi for arms i used to use then for putting 4" ports in my sub enclosures it would work wonders for this

I remember seeing a picture of an ICQ's friends glove box. It was a acqarium that had a plastic bag (clear) taped over the top so that you could manage the equipment in side. I never heard details as to success. Why is it that people want to add fans, etc? Box's that seem to have no air exchange work just aswell.

Just my two cents at the end of this long post...but I say screw glove boxes. I work under less than optimum sterile conditions with my agar....zero contams after making a dozen plates, and just to be on the safe side I mix in H2o2 in with my new agar batches to be colonized by new mycelium slices, works like a charm and a lot less effort. Who knows I may be just a lucky sob...but it works great....cheers

Hehe...I knew that would come up. I don't say that I am super experienced when it comes to agar, and I do expect to have some contams eventually, I just think that it is a lot of work to build these things...of course if I had a tonne of money I would probably buy a pro sterile room for myself. But the way I figure it, getting the odd agra contam, is worth not having to go through the hassle of major construction efforts...hehe. That is the great thing about agar, you know pretty soon if you have contams, and then you can either throw the plate away, or cut a nice piece of mycelium off and start a new plate buffered with H2o2...just my two cents though, I do applaud anyone who can construct a flow hood or glove box...and have good results with it. the problem with peroxidated agar is that you cannot start spores from it. people working with "questionable" prints cannot rely on peroxide- you would either have to start from a live culture (which are hard to come by, unless someone is willing to send you one), or a piece of a mushroom. so, great for cloning and transfers, but the print thing is a drawback. gluck on no contams, i know my house if full of em. youre lucky :)

enter- congrats on the glovebox. its always extremely satisfying to build something yourself and use it :) hope it serves you well, at least til you upgrade to the cadillac :)

i think the problem with carboard glove boxes are that they soak up moisture wich could breed contams in the cardboard.thats just my opinion because i used a cardboard box for a while then i switched to a rubbermaid container with some heavy duty rubber gloves and had a lot less contam problems.

I'm new to agar work and i started without a glovebox 4 days ago.
I took all the precaution to be sterile all the way.
I innoculated the dishes over my gas.(Which is way sufficient for innoculating jars...)in a lysoled area with clean
body (shower) and all.
The dishes were open for at most 20 seconds each and are
stored in an ultra-lysoled rubbermaid.
And i already can tell you that contams are on their way.
On 5 dishes, 2 are surely contamed and 1 maybe.
The rest looks fine.
In conclusion i decided to build a glovebox to decrease the
risk of contams.
Also because it will simplify my sterile process for all my
innoculation now.(jars, grain to grain...) an d because i like to
build my own tools.
It cost me 20$ for the rubbermaid, the gloves, metal rings to
let hands go throught and glue.
I also bought an HEPA filter for vaccum cleaner (20$) that i'll
fix with a fan (free from an old PC )to create positive
pressure inside the box.
The box is not completly air tight but i'm working on it.
Wish i could post a pic....
It took me maybe 1 hour of work +time for glue to dry.
I havent tested it yet but it will be soon.

NICE. your dryer vents just gave me the idea to put it on my own. I made my glove box from a long (about 6', and about 2' high) rubber maid container.. the sides are see through.. and for the top, i used plastic sealed on with bathroom calk.
in the side, there is a hepa filter which was on sale at walmart 59.99, this is sealed around with calk as well.
a flap in the end is where things are put in, and two arm holes are cut.
