Just my two cents at the end of this long post...but I say screw glove boxes. I work under less than optimum sterile conditions with my agar....zero contams after making a dozen plates, and just to be on the safe side I mix in H2o2 in with my new agar batches to be colonized by new mycelium slices, works like a charm and a lot less effort. Who knows I may be just a lucky sob...but it works great....cheers

Hehe...I knew that would come up. I don't say that I am super experienced when it comes to agar, and I do expect to have some contams eventually, I just think that it is a lot of work to build these things...of course if I had a tonne of money I would probably buy a pro sterile room for myself. But the way I figure it, getting the odd agra contam, is worth not having to go through the hassle of major construction efforts...hehe. That is the great thing about agar, you know pretty soon if you have contams, and then you can either throw the plate away, or cut a nice piece of mycelium off and start a new plate buffered with H2o2...just my two cents though, I do applaud anyone who can construct a flow hood or glove box...and have good results with it. Cheers

the problem with peroxidated agar is that you cannot start spores from it. people working with "questionable" prints cannot rely on peroxide- you would either have to start from a live culture (which are hard to come by, unless someone is willing to send you one), or a piece of a mushroom. so, great for cloning and transfers, but the print thing is a drawback. gluck on no contams, i know my house if full of em. youre lucky :) enter- congrats on the glovebox. its always extremely satisfying to build something yourself and use it :) hope it serves you well, at least til you upgrade to the cadillac :)

i think the problem with carboard glove boxes are that they soak up moisture wich could breed contams in the cardboard.thats just my opinion because i used a cardboard box for a while then i switched to a rubbermaid container with some heavy duty rubber gloves and had a lot less contam problems.

i am absolutely inexperienced with agar at this point..ive got some agar and all the shit to do it with just havnt gotten off of my lazy ass..

LO I'm new to agar work and i started without a glovebox 4 days ago. I took all the precaution to be sterile all the way. I innoculated the dishes over my gas.(Which is way sufficient for innoculating jars...)in a lysoled area with clean body (shower) and all. The dishes were open for at most 20 seconds each and are stored in an ultra-lysoled rubbermaid. And i already can tell you that contams are on their way. On 5 dishes, 2 are surely contamed and 1 maybe. The rest looks fine. In conclusion i decided to build a glovebox to decrease the risk of contams. Also because it will simplify my sterile process for all my innoculation now.(jars, grain to grain...) an d because i like to build my own tools. It cost me 20$ for the rubbermaid, the gloves, metal rings to let hands go throught and glue. I also bought an HEPA filter for vaccum cleaner (20$) that i'll fix with a fan (free from an old PC )to create positive pressure inside the box. The box is not completly air tight but i'm working on it. Wish i could post a pic.... It took me maybe 1 hour of work +time for glue to dry. I havent tested it yet but it will be soon. Cheers Duss69

NICE. your dryer vents just gave me the idea to put it on my own. I made my glove box from a long (about 6', and about 2' high) rubber maid container.. the sides are see through.. and for the top, i used plastic sealed on with bathroom calk. in the side, there is a hepa filter which was on sale at walmart 59.99, this is sealed around with calk as well. a flap in the end is where things are put in, and two arm holes are cut.
