Help with Poly-fil.

Sorry for the cross-post, but I haven't recieved a satisfactory answer,

I want to put a poly-fil filter on top of my 1 pint wide mouths. How big of a hole should I drill? How tight should I stuff the poly-fil? Last time I stuffed it as tight as humanly possible and it didn't work out for me. Saying that I need to stuff it "tight" doesn't really narrow it down for me.

I use a pencil sized hole and I stuff it in as tight as possible. Actually I pull it through until it is a snug as its gonna get. You still need to pressure cook or boil and a covering of foil is a must.

here was my first attempt:

I used 1/2 pint jars. Made a hole about the size of my pinky (Slightly larger than a pencil) Stuffed as tight as humanley possible with poly fil. After innocing the plates, nothing happened for like 2 weeks so I forgot about them. Went back much later and found some crazy ass comtams on the agar. Like pink, brown, orange molds. It was crazy. Oh btw i did PC these agar plates before use.

i dont think agar really needs air exchange.

Ive found that poly fill does wonders in quarts. but i dont really see the need in agar. ive forgotten about agar plates before and found that they almost always contaminate eventually, i havent had a plate last more than 3 months by itself.

id try again and just use normal lids. save the poly fill for quarts or half pints when they need to breathe

Ahhh, I see. Agar halfpints. Yeah, you probably could have skipped the polyfil for those. But they would stall eventually if the jars were perfectly airtight. Was the polyfil covered with foil? The fact that it took over 2 weeks for something to appear probably means something got through the polyfil. Contaminate spores most likely cannot penetrate the polyfil directly. But if they land on it, they will germinate and press their way through like a tree root through a sidewalk. You must shield the polyfil from falling spores, this goes for filter disks too. Either store in front of a running hepa filter or cap with foil.

Is Poly-fil nearly the same texture as cotton? I tried some cotton today in 3/4 in holes on 1/2 pint jars. I pulled the cotton through the lid, covered w/ foil and PC'd. When I went to innoculate I found my needle pushed the cotton right through my hole. It sounds like I made too large of holes and didn't pack the cotton tight enough. Afterwards to correct my blunder I placed non-stick sterile gauze pads over the holes and taped the gauze. I hope the gauze acts as an adequate barrier.

polyfil feels like polyester, not cotton- i think cotton may harbor water more easily than poly will... poly doesnt feel like it absorbs water, while cotton very easily wicks it up.

instead of using one hole for polyfil and innoc., you can stuff one hole w/ poly and create another, smaller hole for innoc. only to be taped after innoc. innoc. in a glovebox to reduce the chance of open air contams. just easier and also you dont want to breach your poly barrier yourself (ever had it fall in on you? sucks...) if youre using 1/2pints for pf tek, why not just go verm barrier? or are you using the 1/2's for agar?

I did go with a verm layer. I thought a Poly-fil type set-up would encourage good gas exchange.

yer not supposed to inoc through the poly fill hole.
and no, poly fill filter floss is not cotton.
polyester strads, and cotton are different :smile:

if you are doing pf, why dont you have another hole with tape over it for inoculation..
you arent supposed to be able to inoculate throug hthe poly.
hehe
