Ultrasonic vacutainer spore mixing - A power technique suitable for anyone

Today, I tried the ultrasonic spore mixing technique I had heard about. I had a spore solution in a vacutainer. Immediately after I made the solution, my compound microscope revealed the spores were clearly visible in the solution with minimal sticking to the walls. In only 8 hours, though, I could see that many spores were stuck to the walls of the test tube. Shaking the vacutainer violently dislodged many of the spores from the walls but there were still more on the walls that in the solution. I can imagine after many days, the spores would mostly stuck hard to the walls, but I don't have evidence of that.
I opened my ultrasonic humidifier and turned it on then carefully put the test tube across the ultrasonic water spurt, moving the spurt across the length of the test tube slowly (see note below). You could tell the ultrasonic waves were easily reaching the inside of the test tube as the inside water was spurting and slightly bubbling from cavitation. I then re-examined the spores under my microscope, again through the test tube.
Much to my astonishment, the spore dispersal was perfect! Spores had entirely been dislodged from the walls of the test tube and the concentration floating past in the water was extremely high! This technique works very well to mix the spores, much better than violent shaking even. I don't know if these waves could pass through plastic to the inside of a spore syringe too, but they passed through this glass test tube like there is nothing there and did a great job of mixing the solution.
Note: If one does this, be careful to not get your fingers deep in the spurt as this will drive water through your skin and create a very painful sensation.

id be careful if you try it with plastic. i was playing with one of my ultrasonic foggers and was melting holes in plastic like it wasnt nothin.

I did that once but didn't put my finger too far down, but it was still a little painful :/ lol.. I did it a couple more times after that.

Yeah, probably if the waves don't pass through, they'd be absorbed in the plastic which would heat it up really fast like that.

I put my finger WAY down there... once. It felt about like someone sticking a knife in my finger. I was amazed at the apparent lack of damage. Being so painful with little real damage, it might be a good torture technique or something.

TLG has been treating their syringes ultrasonically for awhile now, and from what I remember, there definitely is a noticeable difference. A small ultrasonic parts cleaner from Ebay is perfect for the job. Load syringes/vacutainers into their, and go.

 Yup, they work GREAT Mine will hold about 50 syringes at a time. Or 100 tubes.
