Lc questions among other things
    #4902587 - 11/06/05 10:39 PM (18 years, 6 months ago)

ok i searched all over the place and cant find the questions that i am asking and if it is on here plz just post a link so i can research it and not bother you guys........
ok for lc can i just take a small piece of myc from colonizing poo and put it in the jar with malt and water ? (like a piece the size of a dime or somethin) instead of using spores?
and also for wbs i have a serious problem with contams (i live in a house that was like built in 1920's or so ) can i soak the wbs and then simmer for 30 mins then pc ? or would that be too much ? i do appreciate all the answers that you guys will give me ( or a link is cool too thanks again guys)

ive read that you can use myc to make liquid cultures, havent tryed it yet though.
thats how i do my wbs. soak over night, simmer for 20-30 mins, drain till it doesnt drip anymore then load up the jars and pc for 60 mins.

or... steep your grain for an hour.
Load your jars and PC for an hour.

If you add grain into a LC jar there is a greater chance of contamination, from posible endospore still inside the grain. But it could be fine.
A better plan would be to just add a small piece of mycelium.
It doesnt take much, the smallest little piece can fill a LC jar with fluffy mycelium in short order.

I beleive he said taking a piece of mycelium from colonized poo...not grain.

To answer your question, yes it will work.

Spores, other LC, or live mycelium will inoculate an LC.

After putting a chunk of myc in there, I'd also throw in an agitator (marble or piece of glass or something) to break the mycelium up when shaken.

LC's are very prone to contamination, if ever exposed to open air.

You can introduce a piece of mycelium tissue into a contained LC, in order to create a large liquid culture. However, the tissue must be clean & the procedure done in very aseptic conditions - like a glove box. Otherwise - when you open the LC to place the tissue inside the container, the LC often also becomes contaminated.

Best procedure I know is to use a BIOPSY needle to extract clean tissue from the inside of a mushroom stem, then inject that tissue into an LC. That procedure eliminates most exposure to open air.

Yes, I have done it with spore syringe needles. Get a thick stem and wipe its sides with alcohol/bleach or cut it, then pass the needle all the way through it, it cuts out a core of stem. Make sure the syringe has air/water in it so you can inject it into your LC into a self healing silicone port.

Best bet for local finds is feed & seed stores that cater to farmers, horsemen & cattle / cows.
10 ml / luer lock / 18 gauge / 1.5 inch needle.

I think a couple vendors here - sell them - also.

On line -- there are many sources.

Those same feed stores could also have the needles in 14 gauge as well.
The 14 gauge are great for liquid culture syringes, and biopsy.

just take some boiled water, put it in a baby food jare( with hole and foil on the lid after youve steamed it a while), about 2wice the amount of water for one syring,(let the water cool of course) take a chunk of myc with steril spoon (ive used my hands w/gloves) put it in jar, shake with ur finger over the hole (remember where it is), extract with sterile syring from the hole in the lid of the jar. and yes i would suggest an agitaor liek glass, ive had nothin but success, use the oven also and try to get everything as steril as possible, i would think poo would make the water fairly dark tho and you cant see how much myc ur gettin in but i could be wrong
