Incubating chamber problem
    #4826381 - 10/19/05 06:02 PM (18 years, 5 months ago)

I just went out of town feeling pretty good with the construction of my incubating chamber, thinking I'd have pretty well colonized the jars by my return. I had water and and fish tank heater keeping the tank at 83 degrees. I built an slanted condensation remover to keep the water from falling on the lids.

I believe I may have killed them since they have changed from bright white to sandy color and if I were a bettin man I'd say it was the humidity that did it. BooHoo.

One jar has the black fungus in it, that was sad to see. Are the rest of the jars crapola?

I will put together a dry incubating chamber with a regulated heating pad instead of the water.

Thoughts are appreciated.

Kindly,
hohum ... more hum than ho at the moment.

I'm a little confused. The humidity shouldn't have much to do with anything because the jars are sealed, right?
Could the problem have more to do with air exchange? Are there holes filtered with tyvek or polyfil?
Could this sandy color simply be from mycelial waste? If you could post pictures, that would be of great help. good luck.

it wasnt the water that killed them...sounds like you were just unsterile...black mold does not come from wet heat i mean...its hard to tell without pictures and not a very good description...

Hmmmm.... well I thought there might be too much humidity because there is some rust on the threaded metal part of the some of the lids and the temperature has been theoretically perfect by the research I was able to dig up.

Only one jar has the yummy black goop the rest just turned from White mycelium to beige and seemed to stop growing. No green or black stuff just what looks to me like mycelium that have been soaked and have darkened as a result. Fact is, I don't know. I will post a pic or to for y'all.

Yeah dude, that rust doesn't matter. I've experienced that before and it didn't lead to any problems, besides making the bands on the lids difficult to twist open. but a little elbow grease fixed that problem...
but if you could post a pic that would be most helpful for diagnoses.

Here are the pics. Notice if you can the darkish rings around the growth, which has pretty much stopped and the white balance is a bit better in the second and third pics, but unless I am crazy they are not really as white as they were before I left. They have beiged.

So thanks to the wiser folks than I who might shed some tips.

not sure if the pics will automatically insert. In case they don't...

It may be possible that growth has simply stalled. Often times a change in temperature is sufficient to kickstart it back to life. Try removing the jars from incubation for 12 hours and then put them back in.
Hope this helps.

Plus, can't tell from the pic.
Do you have holes drilled allowing for air exchange? Air exchange is key for good growth.

Yes there are four holes on top of the lids where I innoced the jars. I have removed the tin foil lid on the aquarium and put a towel on top to allow some air exchange in the aquarium and to keep excessive condensation down... I am encouraged that it might only be a stalling issue.

Interesting:

I have had a jar with the black contam which I discarded just a moment ago. I thought it was very interesting that while there was only partial coloniztion on the outside of the cake where it pressed up against the glass, the caked seemed mostly colonized on the inside. Quite a bit of white mycelial growth occupied the cake and it was very dense; I had to use a screwdriver to break it up. I would have assumed that there would not have been that much colonization since there were only smaller circles of colonization on the outside of the cake.
