Question: Laminar Etiquette

Advice needed.

I am a vetaran grower who has always used glove boxes for sterile work, with much success. I have recently built myself a HEPA flowhood and realized that I had no experience with one before, and I have several questions for anyone who does.

Is there some simple algorhythm i can use to determine the area of "clean" space in front of a filter (I have a 12"x24" with a 495CFM fan)?

Should I feel a solid flow of air from the filter, or is it actually meant to be gentle, and is gentle still sufficient to retain sterility?

Does having a LFH mean I should feel less paranoid about careful sterile practices, or simply that I now have a larger clean workspace to utilize and more freedom of movement?

I should mention that the idea of opening an agar plate in an open room terrifies me, and is the kind of thoght that keeps me up at night.

Any assurances/advice would be most appreciated.

thats a good question?? i used to work at this place making the silcone diaphrams. for navy suit controlers. i worked under a flowhood and i felt a steady but slow suction. you could only feel the suction like 2-3 inches away from the back wall.

Continue to follow scrupulous sterility techniques. The flow should be firm, but gentle. No wind tunnel here. You do have a larger area to work in and more freedom of movement. I suggest that you put a large see-thru hood/chamber around where the filtered air comes out. This decreases any particulate matter from floating in from the top.
I can't think of any formulas. sorry.

You should work very much in fron of the filter, as near as possible,perhaps 5-7 inches away.
There is a simple test for how far the laminar flow goes in to the room.
Light a cigarette or an incense smoke stick and hold it infront of the filter. You should be able to see the form of the air stream this way and be able to tell how far it reaches.

Some of the rules of laminar flow are different. The flow can blow contams from your hands or clothes so stay down wind from your work. Also let the flow run for about five minutes to clear the air before use. Flame sterilization of tools does not work well with laminar flow because the flame disrupts the flow so either keep the flame outside the hood or use an electric incinerator. Laminar flow is great when it works. You can test how well it works by exposing agar plates to the flow.
