Or one can just put the needle tip in a flame and pull back the plunger.

i sure wouldn't rely on that. flame doesn't instantly kill every microbe sucked thru it. try that and shoot the air on some agar... also flame is inappropriate for some situations, such as a lysol-laden glovebox.

steam is far more effective than dry heat. pc it, steam kills everything inside. baking it hot enough to do that would ruin the lid's rubber seal. proly just a spoonful inside is plenty.

this seems like a lot of trouble.
i liked what you did with the store bought flavored water for lc use. ingenius.
what use does an airport serve if you have to go to so much trouble just to keep up with the maintenence of oxygenating your jars?
in this case instead of trying to get "sterile air", wouldn't it be easier to just use a polyfil filter for your jars?

it's no great trouble to squeeze out a blob of silicon or two. it's no trouble to suck air in a syringe. i'm not trying to sell you an air kit, if you don't want to try it, don't. someone somewhere will, and that is good enough reason to post it. see this thread for current example of trouble some folks have with polyfil- wet polyfil one last thought- the easy way isn't always best. polyfil can get wet and let contams thru. esp. when doing liquid cultures. this air supply allows one to give LCs a hard shake [try that with a polyfil filtered lid] to break up mycellia, inject sterile air. one LC can make many syringefuls of inoculant, making the 'trouble' rather worthwhile. some folks have trouble finding polyfil, maybe they live in thailand in a hut on edge of jungle with no walmart in sight. maybe they just want to experiment with something new to see if it can be done.

speaking of which- that culture is now 6 months old and still doing well, just filled another syringe of mexA tissue a few days ago. growth has never gotten dense but there's only a 2% sugars content. drawing the LC out of the lower airport works like a charm as gravity settles the tissue to the bottom so you can suck it right into the needle, air bubbles in the upper airport to compensate the volume lost. i'm seriously thinking about trying to 'feed' this LC by injecting a little fresh karo water.

Heres an airport I made.

Its an air valve from the plumbing section that was a hell of a tight fit but threaded onto a polyfilled syringe so that the airpump plugs right into it.
I Pc my airports everytime I use them is this necessary?
