blackout said: Agars jars work well but if you have say 20 jars on the go then you need 20 caps to be built. Using filtered syringes you simply poke 2 holes in each cap and but a blob of silicone on them. Then use just the one filtered syringe. I put bleach in my syringe and squirt it through the filter before putting it in my jar.
true enough, hard to get much simpler/easier/cheaper than stuffing an old syringe with polyfil, poking a hole in a lid with an icepick and applying a blob of silicon adhesive. that's it, do it for a penny a jar, nothing permanent so jars can be stacked anywhere. lay jar on side, upside down- matters not. shake a LC until the tissue fragments, no need for glass or magnets. but one big advantage- you can place the silicon on a plastic bottle never opening it and slide the needle in to inoculate. you could inject apple juice into a gallon jug of distilled water, right from the unopened jar of juice to the unopened jug of water to create a nutrient solution then inject a few spores- voila! a heat-free flowhood-less way to make a gallon of liquid inoculant at a time for mere pennies in just a few minutes wih very few supplies needed.

here;s another thread to check out- http://forums.mycotopia.net/showthread.php?p=194915
one thing the airport system makes feasible is 'canning' nutrient solution the old fashioned way- lids with siliconed holes, rubber-side down so it seals fully as it cools. once cooled/sealed inside the jar will be a partial vacuum. if carefully inoculated to preserve most of the vacuum, his experiment seems to indicate that mycellia grows faster in the low pressure environment. this is still bleeding edge stuff with a need for more evidence/confirmation but if the phenomenon proves real/consistent/significant it could change how we do LC's, dramatically speeding up the growth of tissue in liquids. worth a few more tests, imho.

Yes, I was talking to you about the apple juice at mycotpia. You can easily put a needle in the perforated hole on the tetra pack. Juice is usually 10-12% sugar so add 2 parts water to one juice, though weak LCs work fine too. Some sports drinks are now 3-5% sugars instead of the usual 10-12%. Lately I have been working in a/c bags as a cheap glove box. If you made a jar of 50/50 water/honey and heated it up it will be fluid enough to pour into a large bottle of water in a glovebox.
If working with vacuum just make sure you dont lose your whole syringe when you inject.
I do most of my sterilizing in another house, so transporting LCs used to be a big problem for me, it does solve it.
I have had great success adding large amounts of LC to grains and have some projects I will be working on soon that need a lot of LC.

I just use one of these uncommon items (Socorex auto-loading syringe). Slide the feed tube into a LC rubber tube, agitate jar & pump away. Makes it EASY as eating pie, to inoculate 40 or 50 spawn bags, or jars - at one setting.
where did you get that gun???!!!

eBay for $26 plus S/H. They run arounf $140 retail & depending on what fittings you get with it. Soccorex is the best in the world.

In the picture with the airport being used to pull sterile distilled water... was the water used to make syringes or straight for liquid culture use???

it was used to make syringes in that particular case. the bottle of fruitwater was used as a liquid culture [mexicana A]

so was it, pull water from bottle, and hydrate spores with a brand new syringe?
if so, how was the contam turn out?

no, the print was placed inside small ziplock bag and water was injected, stirred to mix then sucked back into syringe

permanent endless supply of sterlie air sometimes one needs a source of sterile air. here's a simple way. quart jar fitted with twin silicon ported lid about 1/2 inch of water in bottom run the pc to sterilize insides then place airport in one hole to equalize pressure. empty syringe inserted thru other hole can then be filled with sterile air as needed. the polyfil syringe will filter replacement air maintaining equal pressure
