Are fully colonised agarmediums prone to contamination?

Can you for an example add pasturized h.poo on top of the colonised agarmedium in order to get first generation spawn?(I use jars not petridishes)

Well, just keep in mind that there are still lots of unused sugars underneath the top mycelia layer.

The beneficial part of spawning poo with grains is that you get multiple inoculation points all throughout the spawn, while with the method you're proposing you would get one inoculation suface, while the rest of the poo would have time to ferment while the mycelia worked its way through.

The advantage of having first generation spawn would be outweighed by the extra likelihood of contamination, so I'd advise just using the agar to inoculate grain, and then take that to poo.

If you have extra cultures and poo, though, no harm in trying it out!

As said, the underside of the agar needs to be in a sterile environment, thus it is used to inoculate sterilized jars of grain. Once the grain grows out, you can use it to inoculate the horse manure. If you attempt to inoculate manure with agar, it is almost sure to contaminate.
