Do I have to make a spore print?

or can I make one from a little slice of fresh mushroom tissue? That would save me a lot of time. Every tek I read on LC doesn't mention inoculating the malt solution with anything but spore solution...

from my reading, i only recall using tissues on agar...I may be wrong.......

look into cloning. check out anno's site. fungifun.com of something like that.
but, i would recomend a liquid culture...

The nice thing about spore prints- is that you can make a shit load of them. If you have some contams you can just use another print. The other good thing about it is that in a year from now you can take a print and grow from it. With a dish- they are perishable. I have used syringes as old as 2.5 years old.

Yes you can make a LC out of fresh mushroom tissue - this is called cloning. Cut a peice of tissue from the centre of a stem with a sterile knife and drop it in to your LC in sterile conditions.
Alternatively, you can take a spore print and start a LC with that - instead of mushroom tissue, just use a sterile knife to scrape a few spores into your LC in sterile conditions. This is called multispore innocluation.
Take a few prints and store them in little plastic seal bags and they will last for many years. I have had no problem starting a LC with a three year old print.
