As has been discussed above, the "nitrogen content" (at least, in any basic sense) of the substrate is a misleading number. Excluding water, nearly every molecule which constitutes the fungus contains vastly more nitrogen than a psilocybin molecule does. Even speaking only of proteins, which make up 30% (ish) of the mass, each protein molecule contains several thousand times more nitrogen than a psilocybin molecule.

This is the whole point of this thread. Nitrogen content alone has no impact on alkaloid production. The question, as stated several times above, is what form the nitrogen must be in in order TO have an impact.

As I've explained in other threads, there's little reason to think that trytophan precursors will get you very far in increased alkaloid content, because tryptophan decarboxylase is inhibited by tryptamine, psilocybin and psilocin. As a result, there's little hope for coercing the production of large quantities of alkaloids by increasing tryptophan levels over those which naturally occur.

Needless to say, none of this is true of compounds which metabolically occur after tryptophan.

I really don't have time for a proper post, so just quick hits:
Ammonium succinate is expensive. It can be easily and cheaply synthesized from succinic acid and ammonium. I would consult the proper "Current Protocols in ... " series for hints of why that particular N source was chosen. In the very least it should provide pointers for further research. As for tryptophan decarboxylase - make sure the data applies to Psilocybe's enzyme, which I would suspect might be evolutionary convergent (analogous) to those of plants, and thus could be subject to different regulation (also, some inhibitory regulations can be overwhelmed by excess substrate). Since the biochemical pathway to psilo. isn't well delineated, one would have to rely on empirical data anyway. But I wouldn't extrapolate the paucity of empiricla data we do have - in this case, beyond 11 days and liquid culture. Limiting ourselves to that offers interesting enough venues.

Use of different nitrogen sources by the edible ectomycorrhizal mushroom Cantharellus cibarius

Abstract The growth of three strains of Cantharellus cibarius on liquid media containing ammonium, nitrate and bovine serum albumin (BSA) in different combinations was determined. The most readily utilisable source of N was ammonium. BSA utilisation was limited compared with media containing ammonium. Growth on nitrate was also poor, suggesting a limited capacity of C. cibarius to metabolise this nitrogen source. There was some indication of considerable intraspecific variation within C. cibarius in the utilisation of nitrogen sources.

Possible links between atmospheric nitrogen deposition and the observed decrease of sporocarp formation by C. cibarius in Europe are discussed. We highlight the potential ecological significance of bacteria associated with C. cibarius which may circumvent the need for fungal extracellular enzymes to access complex nitrogen sources.

Ponder that P. Cubensis is historically referred to as a "DUNG LOVER".

There is a lot of data (Penn State - for instance) that a compatible Nitrogen content, all else being optimal - under ideal conditions results in optimal crops.

Stands to reason, optimal fruits - give optimal (natural) potency.

High "N" H/poo or h/poo / dung based composts have given me - what I would consider optimal (natural) potency.

Nothing but a lot of grows - over a lot of years - to back that up.

But, that is enough for me.
