Ok, if you're trying to isolate a single genome strain, at what point should you take it out of incubation? When the mycelium is completely covered over the agar or when the growth is significant enough to cut out a nice peice? Any help on this would be greatly appreciated...Thanks!

If you are using the mycelium as an inoculant, then just cut a small piece out for that purpose, then incubate the plate until the remainder is full. You can store the plate in the fridge for future use.

it would take a bit of work to isolate out a single genome as you suggest. you would pull pieces from the original plate and let them grow out on another plate and then continue that process until you are getting the myc growth that you desire. something like that anyway.

Ah ok, so the first dish wouldn't have to be completely covered, just enough so that I can pull some myc off and put it in the next dish...right?

Yeah eric, you got it.
My aim when i use agar is to keep isolating till i'm happy with what's on the newest plate. Then i inoculate a liquid culture with my new isolated strain.
