I'm using the PF TEK method, and while it seems that most of my jars are doing okay, it seems like the colonies are getting to a certain point, and not getting any larger. There is one jar in particular that began as my best jar, far outpacing all the others, but the colony hasn't spread in about a week, and it only covers one side of the jar. I've been poking around for reasons for this, and the only thing that it looks like I can do is remove the tape I put over the holes in the top of the jar.

Is this really a good idea? Are there any risks involved? Are there any better ways to speed up colonization?

Do it. It should be fine. There's really not much need for the tape anyways. Try using Tyvek.

That is IMHO the biggest flaw in the PFTek. Not so much the use of tape but the fact it isn't removed immediately after sterilization. After PCing it is not needed and it can only interfer with gas exchange once spores germinate leading to stalling of mycelium growth later on. Your verm layer is all thats needed as far as a comtam barrier, I myself recommend using foil right from the start, you can write on it it helps maintain proper moisture and gives an additional sense of security. GL

i usually never left the foil on pf jars either and never had any problems. that dry verm layer has always done its job.

I agree, that is a very big flaw.....but all I do is PC the jars with the tape on, then remove the tape for innoculating, and keep it off from there on out. I don't use foil or anything.

The dry verm layer is enough to keep out contams.

If your jars are stalled, try removing the tape, and flipping the jars on their lid for a day or two. That should help start it up again.

Sounds like the reason your jars have halted is because they can't do gas exchanges because of the tape.
