Hello,
I was curious as to what the effects are of taking one strain of mushrooms and growing it out through many generations? (I'm no biologist.) What effect would this have upon the genetics? For example, what would the difference be between the first generation and, say, the 20th? Smaller gene pool? Less variation..?

Any input is greatly appreciated.

When talking generations in mushroom culture, you are really talking about the number of cell divisions that the mycelia is going through. As far as I know each cell has a finite number of divisions that it can go through before mutations and degeneration set in. Most of the time, a culture is started, expanded onto agar, and from there, if it evaluated as a "keeper" a small portion is transfered (or a few) to agar slants for storage - this keeps the stock culture very close to the original cell divisions, and it is easy to expand and will grow vigorously.
As it goes down the line from the spawn producer it is usually expanded 3 times before being sold to market, so commercial spawn is usually 2nd or 3rd generation, unless a customer is specifically requesting 1st gen spawn. After the third generation (you've expanded the mother culture into 10 master jars- G1. Then you've transfered the masters into 1,000 filter patch bags- G2. Expand the bags to 10,000 G3 spawn bags for sale) spawn bags are made, they are expanded to bulk substrates, all this causing massive cell division in the original culture, but it's still totally within commercially productive parameters.
Once the mushrooms fruit from the bulk sub, they are taken to market. At this time, the original culture has one through so many cell divisions that it reaches a sate called senescence. It now is less likely to be good for cloning, as further cell division would lead to poor yields, misshapen fruits, disease, and general mutations that are to be avoided.
So the generations are limited by the number of cell divisions that you go through.
Everything you're using genetically should be a transfer from your original master plate.
