Your jar filter prep

Hi

Was just wondering what some of the experts do for their filter preps. I myself use jar then coffee filter then lid then screw on ring thing that holds lid down, then breathable surgical tape over innocc hole then tinfoil then pc and yea... I've been having major problems getting a honey water culture going (always start to smell sour  :confused:) Anyway was wondering if it was my filtration method??? Any hints or tips? What do you guys do?

IMO...fuck all that taping, and discs and what not.Its all about polyfil ended tubing and self healing injection sites.

Could it be contammed spores? As long as your filter is a suitable one, I don't see why it would be the problem. If you are sterilizing your LC properly and using proper sterile precedures during inoculation, it could be the spores.

I was thinking that... but the wbs jars i've done using the same method and spore syringes and all turned out fine... The coffee filter does get wet though cause it's on the inside, maybe thats a prob? It's rather annoying  :frown:

coffee filters are not microfilters, contams can readily get through them. mainly they are used as dust covers on the OUTSIDE of the jars, thats it.

you should switch to polyfil or tyvek they both will keep contams out whereas the coffee filter will not.

Yeah, like I said, "suitable" filters. If your coffee filters are under the lid when you PC them, they will probably get wet and be contam prone. Check Agar's posts. He has a real good coffee filter tek he uses on his jars from what I remember. I'm not sure what your access to Tyvek or polyfil in South Africa is, but I prefer those myself.

Tyvek is unheard of and so is karo... then again so are 1/2 pint canning jars, oh and rye. Yea I need a filtration method... perhaps get that polly fill stuff out a pillow? Then stuff it in my innoc hole? Any other names for Tyvek? or anything else I can try? Thanks for all the help guys. :smile:

If you are unable to acquire tyvek in S Africa I could send you some, pm me if you need.
Polyfil is used in pillows as the stuffing so maybe you can get some out of a pillow.

I used one of our sponsors filter disks that are cut the to the shape of the jar. Drill two holes in the metal or plastic jar top. I just poked the needle right through the filter material and did not worry about it. I also jammed polyfill into the holes. Nothing on top of the jars, but you could also put a coffee filter over the jar and secure it with a rubber band.

19 for 19 on that method so far...

How bout this... Two holes, each stuffed with pollyfill then breathable surgical tape stuck on each end of the hole holding the pollyfill in place. PC, inject, then more surgical tape over the innoc hole after injected. Then coffee filter on top held by rubber band??? Tyvek is unheard of where I live. :thumbup: :thumbdown:
