Can someone explain grain 2 liquid transfers in detail?

Heres the procedure im considering. PC syringe for 20 min w/ karo water, wait to cool. inject 10 cc karo water into jar, swoosh it around in jar and suck back into syringe. is it really that easy? some questions arose in my mind while pondering this tek.

after the procedure, the colonized cake will likely be sitting in excess water as it would be difficult to remove all the fluid. Could this cause a contamination if the cake was left to sit for days in this? If you opened the jar and dumped out the fluid, exposing it to contams in the air, and then sealed it back up, would it remain fine? Would you let it sit in the water or expose it?

Does mycellium have to be visible inside the syringe? or it it microscopic like spores? it will be visible in a few days of syringe incubation hopefully...is incubation even necessary?

The colonized cake is solid like a brick...are you supposed to break it up into small kernels inside the jar before doing this tek or does the water sliding along the surface pick up enough myc?

I might try a g2g as well, both sound like fun experiments.

mycelium should be visible inside any liquid culture.
excess water in your jars is a bad idea, and is a great vector for contams to move in. opening the jar and trying to drain the excess water is even more likely to creat contamination.
incubationing a liquid culture is not totally necessary but it is helpful for faster growth IME.
have you thought about just making a karo jar from a multispore injection instead of an individual syringe? i've worked with liquid cultures many times, but the method you are describing seems like you are going out of your way to make what is basically a simple procedure difficult.

so you do it right before you plan on fruiting, cool beans.

i made the LC from spores and i think they all got contaminated from the filter. will know shortly as their fluids have been transfered to a medium that allows contams to be more distinguishable.

the first method you propose is certain possible, but better not to mess with your existing efforts. when you are ready to fruit, pick a small piece of grain off the cake, dunk it into hydrogen peroxide and carefully pop it into a karo/lc jar (preferably inside a glovebox). you can then suck out plenty of lc from that jar once growth begins.

The cake will absorb alot of the karo/water. You won't even get all 10cc's back, so I wouldn't worry too much about there being excess fluid in the jar. Also, if the cake is fully colonized, it will be pretty resistant to contams even IF you had to open the jar up to drain fluid out.

Grain to liquid transfer in detail.

1. Make a LC and let it cool (once you have a colonized jar in your incubator of course)
2. Create a glovebox, flowhood, or clean a small room with no carpets like a bathroom to make it a cleanroom for your work.
3. Go into the cleanroom (thats where I do all my sterile work, in the bathroom) with the colonized jar, and the LC.
4. Unscrew both lids but dont remove them
5. Use a spoon you brought in with you (after you wipe with alcohol) and get a small tiny tiny peice of mycelium from under the dry verm layer (if using cakes, if using grains, a single grain will work fine).
6. Then quickly, lift the lid off the LC, and drop the bit of myc in it (being carfull all the time to not move to much and create airflow, all a/c and heaters should be off, windows closed ect)
7. Close the lid up and incubate for 2 weeks, shaking and swirling the LC every 2 or 3 days.
8. When you got a big cloud in the jar, suck up all the solution into syringes.
