Grain transfer to Liquid culture

Is it feasable to transfer grain to form a liquid culture. I would be using colonized WBS into a gladware like that used in the microwave karo tek.

My only concern would be that since LC's are so easy to contam that opening the wrap would pretty much ruin the LC.

Yes. To prevent contams you could inject some water into your colonised WBS and suck it back out drawing out some myc with it, then inject into your LC.

Build a glovebox if you plan on doing these type things. Doesn't have to be fancy, just a still air glovebox will help out a lot.

No doubt on the glovebox. I have not had a single contamn in LC, Agar cultures or G2G. And let me tell you something. I suck at them. I am clumsy as hell. I'm always dropping shit and forgetting to sterilize something. Like I will open the lid to an petri dish, pour the agar, then put the lid back on...crooked enough that when I jerk my hand accidently it flies away and lands upside-down, nocking open a few other dishes while it's at it...

And of many stupid things like that, I still haven't had any agar plates go bad. And LC is cake compared to agar.

Build a cheap ass glovebox. Buy yourself as clear of a tub as you can. Go to Home-Depot and get one of their pre-cut squares of plexi-glass like plastic that is slightly smaller than the lid. Cut a square in the lid that is smaller than the plastic you got from HD. Lay the plastic flat, get some duct-tape and tape the edges. Now you have a nice clear window to see through

Cut two holes for your arms and you are set.

To upgrade the box, use that hose from the back of the drier (okay buy a new one), and cut it in half. Stretch a pair of kitchen gloves over the end, and stick them through your holes. Tape. Now you got a sealed glove box.

To make it a beast, buy a hepa filter and run another section of the hose from it's output to a new hole in your glovebox. Tape. Now you have a sterile lab.

There are MANY teks to make a box...thats the general idea
