innoculation and sterilization

I really don't understand why such serious precautions are taken during the innoculation phase like wearing a mask and lysoling the place, but after that it is not so serious like the jars are exposed to air and contaminants if it has holes drilled in. Why is t so important that during those few seconds that everything is so sterile but after that the environment doesn't have to be so clean? Thanks

Once you inoculate you cover the holes with tape or foil.
So no air goes in it.
While you incoulate, instead, air may come in thru the holes and lots of bacteria may attach to the syringe which goes inside the substrate.
