how to sterilize plastic baggies?

Hey everyone. Just a quick question that I havn't been able to find anywhere yet.

When storing spore prints or making syringes, baggies are used. But of course, they are not sterile after you buy them at the store. So I was just wondering how one might go about sterilizing them in and out prior to use without melting them.

The best idea I have come up with is swish around some alcohol inside the bag prior to use and then let dry (and let bacteria build back up).

Any ideas?

usualy they are sterile from the manufacturing process, the inside anyway, if you
insist on doing more then Isopropyl Alcohol is a safe choice, just allow plenty of
time for it to flash off

If you really wanna be paranoid first do it with dilute bleach leave it an hour then do the alcohol.

I've never had any troubles from using bags right out of the box. They're completely sterile from the manfacturer.

There is no guarantee that the bags are sterile on the inside. I have called the folks at ziplock and they tell me that the bags are NOT sterile. However, I haven't EVER had any sterility problems using ziplock bags right out of the box. You should be fine without taking any extra precautions.

Theyre sterilized enough from the factory IMO. Were talking about mold spores, they wont stick around there. I've never had a problem either, and I even just fill up the bag with water usually and suck it back up to make syringes from that point.

Agreed. Ziploc can't advertise their bags as sterile in the medical sense, but I've used them to colonize substrate, and never had a problem. They are manufactured at very high temps and come off the line sealed up. They are more than fine for storing prints. They'll be much cleaner than any print you can take that you'll be putting inside.

This is good to know, I was planning on just printing directly into the bags. Just take a cap and put it in the bag, no messing with tinfoil, the one step approach should reduce contams, seal the bag and there are no contams getting in while you wait for the spores to drop. I have no glovebox but will work in a large clear plastic bag, just pick caps with a latex glove and dump them in the bag, no need for careful placing, seal the bag and move them to the centre later.
People can just squirt a small bit of water into the bag and suck up some spores.

I use half pint wide mouth jars with Tyvek replacing the lid:



I sterilize the jars in the PC and just drop the cap at the bottom of the jar overnight. Then I take the cap out and let the print dry up for a few hours. The Tyvek allows the moisture to escape the jar. I think I got the idea from one of Magash's post where he did this with foil at the bottom of the jar. When I need to make a spore syringe, I just take my needle and scrape some off of the bottom of the jar and dump the scraped spores into another sterilized jar. Then I squirt half the water from the syringe into the jar and suck the spores back up. This works best in a glove box, but a Lysol sterilized bathroom will work good too.
