substrate colonization question
    #440310 - 10/29/01 05:25 AM (22 years, 5 months ago)

ok, last night I boiled and inoculated 12 pf jars filled with dark rye flour, everything seemed to go great until the boiling took place, when they finished boiling for an hour (sitting directly on the bottom of the pot btw) I noticed a lot of moisature and water droplets inside of the jars, when filling the jars I didn't pack the substrate at all, I left for a lot of air holes just like it stated in the MMGG. Is this normal? When I inoculated the spore water ran to the bottom of the jar along the inside of the glass. I don't remember this happening the last time I did jars. Could the ratio somehow have changed? I actually opened one of the jars just cuz I was curious, and the dry top layer didn't seem wet, but it was a little moist, not very much though. This is the 2nd dozen i've ever done.

any help is appriciated

You shouldent have left them on the bottom of the pot thats too close to the water
also I wouldent have innoculated the same day you sterilized there still gonna be hot in the middle

I waited 6 1/2 hours before I inoculated, they were totally cool, i'm not worried about that...

what I am worried about is the ratio being switched because I didn't screw the caps on tightly, i'm sure some steam got into the jar in which turned into water. can anyone confirm or deny this? and should I place a dish towel on the bottom of the pot next time I boil? I don't know how else to do it besides letting them rest on the bottom of the pot.

ok my question still isn't getting answered, this is frustrating! one more time...

am I screwed if I didn't screw the lids on all the way real tight? will it leak extra moisture into the jars and throw off the ratio of drf/water/verm? if so i'm just gonna empty the jars right now and throw it all away and start over, no sence in waiting for somthing to happen if it's not going to.

Hard to say. You should be ok- when I pressure cooked my jars I left the lid open with no ill-effects. Just give it time and see if it begins to colonize. Patience, patience.

BTW- I'm not sure what'll happen if you screw the lids on all the way, probably not enough moisture in the jars to break them, but you really ought to leave a little vent I suppose.

yeah I have a lot of water droplets on the inside wall of my jars along the glass, seems like to much water inside. I hope they're ok

i've done this before on a few jars. they will probably turn out absolutely fine, i know mine did. spore water running down the side of the glass is also totally normal.

I feel much better now, could you see the water droplets on the inside after you boiled? they wern't present before I boiled. everything is also very loose and unpacked inside the jar, I was 100% sterile so that's not gonna be a problem. I just think the ratio is to wet, i'm skeptical. I hope you're right.

also one more thing, do the jars need to breathe at all while they're colonizing? I put them back in the box the jars came in and taped the top so nothing could get inside the box from the air. is this ok?

I just had this same problem. Your holes on top of the lids aren't big enough to get enough air, so i just loosened the lids enough so air can get in. No more than 3 days i had germination.

wow, really? did you notice the condensation and water droplets on the inside of the jars after you boiled?
and did you keep them in an incubation chamber? I just have them in the box the jars came in, is that ok?

well this post definatly has me feeling a little better, i'll check the jars on friday (I inoculated on monday morning) and see what has happened. I was pretty skeptical after reading over the MMGG again after boiling, if i'm not mistaken is says to screw the lids on tight before boiling, I thought for sure my substrate ratio had been changed. I was ready to throw the substrate out and start over.

You'll be fine. I used drinking glasses with foil over and the substrate got a little darker and a little wetter, but they are all colonised and going mental. Got shrooms on five of them so far. They had small droplets of water against the glass too, all the way through 'til I birthed them. I don't see it as a problem, but if you're worried then put them in a warm but dry area so that some of the water will evapourate (obviously leaving the lids loose)... I wouldn't recommend it though, as like I said I was okay. But it's something you could try if it looks like being a problem.

Good luck mate, and if you've been sterile I doubt you'll need it.
