Question on cloning
    #4403225 - 07/14/05 01:09 PM (11 years, 4 months ago)

When this batch of mushrooms I am growing now is done I am interested in trying to clone. I have never don this before, but I am trying to expand my knowledge of the hobby. So, My plan is to take a peice of the base of a mushroom, let is sit in 10% h2o2 solution and then place into a lc made from malt extract. Is this correct? If not, what part is incorrect? Also assuming I am doing this correct, how long will it take for the lc to be finished. I know when using spores I wait about a week to use, but I am not sure what the timeline would be like with this. Any help would be appreciated.

First off take the tissue sample from the largest specimen that is in a cluster/group this is always the best choice. 10% may be a bit on the strong side, I use 3% and take a sample from the inner core of the stem chop it up in the h2o2 let it sit from 20-30min. then transfer to agar, I haven't done LC with tissue but as long as you have something in the LC container to agitate and break apart the myc you should do just fine. Your judgement as to a time frame will depend on conditions and how "thick" a LC you want. GL
