Proposed sterilization of PF jar.

Just wondering if this sounds retarded or reasonable. Agar was talking about how you can microwave-sterilize jars and I was wondering if this would work(quite similar to his suggested method): Fill jar with substrate..blah blah cover with some coffee filters, and rubberband them down tightly optionally cover again with ceran wrap(may not be good to steam the filters and keep the water in) microwave with a plate on top of the jar for 2:30 minutes 3-4 times and then just inoculate through the filters
I dont think this is much different from his method, but this is what I was planning on doing and wondered if it seemed practicle. Thanks for any help.

Well, you would need some sort of lid besides a coffee filter, or your sub would dry out. Plus, I wouldn't be too keen on leaving coffee filters under an unsterilized metal lid, that would just be begging for contams.. If you could rig up a microwavable lid, with a self healing injection port, I think that would be your best bet..

what if i put ceran wrap underneath the filters and used them mainly to hold the pressure and keep airborne contams from getting to the innoc holes in the wrap?

I don't think it would stand up to the nuking.. There's only one way to find out for sure.. That's trying it, but steam sterilization is so easy, I don't see why not just do that.. It worls for sure!

well, for one i like to make things difficult for myself. and for two its not too convenient to leave jars boiling in a pot for an hour or two. im at home for summer, and although my parents probably wouldnt be mad or dissallow me to do it, id rather just keep it to myself since this is more of a run through anyway. i dont plan on doing this for realsies until i go back to school where im not putting my family at any risk, but ill let u know how it turns out.

Good luck.. If I where going to do that, I would get a microwavable tupperware, put a little hole in it, tape over the hole, nuke it, lift the tape, and then innoc.. I would then replace the tape, incubate, and when it was 50% or so remove the tape.. I think that would be a pretty slick nuke tek..

how well does microwaving kill contams? i gots so much tuppaware...would be cool to use it

It works real good on LCs for me.. But I've never used it to sterilize anything but LC..

Not very well if you don't understand what is happening in the microwave. Your main problem with microwaving is hot spots and moisture loss. I have been doing a few experiments, it is too early to write up a foolproof "tek" but if there is enough interest in micrwave experiments I will write up a guide as to what has worked for me in the past and what is presently happening. There still seems to be very little interest though...
I have some details in the growlog section on wheat I microwaved for only 25mins and fruited successfully.
I certainly think fractional sterilization is possible in the microwave but I am trying to repeat my single microwave step for grain (with more grain) which would be a good breakthrough.
