PDYA LC question

Here is the recipe used:

-PDYA-
500ml water
5 grams of instant potato flakes(Potato Buds Brand)
9 grams agar agar
10 ml honey (Raw natural pure honey - Beemaid Brand)
1 gram of Nutritional Yeast

This was all combine in a quart jar and PC'd @ 15 PSI for 45 mins. Once the quart jar was cooled enough to touch, it was transported to my lab. Preparation for filtering Ala coffee filter in thru a funnel and finally into a baby bottle retro fitted per Agar's BB tek. (with one exception, a FD cut specific to the bottle)

I get everything  filtering, filled 1 up 4oz. Went on to #2. Left everything to sit for a 1/2 hour to do some menial tasks. When I came back, everything was more gelatinous than words can describe. SUPER thick and most deff unusable.

So what did I do wrong? I thought many ppl made LC using agar agar and a nutritional base, or was I wrong? Any help would be appreciated.

And yes, the recipe was followed and measured completely in lab like atmosphere, lab tools and exceptional care. Kind of disheartening, but then I am all over it : "oh boy! I made a mistake, time to learn from it" Synapses Firing

I had a glass bead agitator added to the Baby Bottle, while filtering I noticed even in a somewhat liquid consistency the bottom liquid was hardening extremely firm. So firm that the agitator was almost fixed an immovable. What went wrong?

The purpose of the agar is to solidify the medium, so it is not fitting for a liquid culture. If you leave out the agar from the recipe, you will get a liquid medium with the same nutrients as the original recipe.

Note that if you filter the medium after sterilization, you either have to do it aseptically, or resterilize it afterwards.

The tater flakes & yeast will get you a milky solution, which IMHO is not a good thing.

I would suggest a 3% solution of karo -or- dextrose and/or light dry malt to water. Karo -or- dextrose is clear. Light dry malt will be a little gold colored. PC 15 or 20 minures.

Yep, if it is cloudy it is hard to see what is happening inside, and harder to spot unusual growth (contams).
Don't toss what you made. It should make some good agar plates, thought usually you would add about 1.5g agar per 100ml.
Use agars recipie or use your honey to make a 2-4% solution

You used my agar recipy for a LC is the problem.

You don't use this recipy for making liquid cultures.

Use the karo tek or a malt based tek for LC's.

I don't know how you got mixed up on this bro.

I hope it wasn't something I said.
