Trouble with the old bag...
    #4355934 - 06/30/05 05:08 PM (11 years, 9 months ago)

I'm going through my first trials with mycobags and success has been limited.

All four were noc'd with a known healthy/clean LC, about 5cc's of what I could draw into the syringe each. (forgot to add anything to jar LC to shred myc.)

The first two contamed, I think I (way) over filled them.

Second try, I noc'd two bags (much less full) and a 1/2 pint BRF jar as a control for contam/growth speed.

The jar is 95% to fully colonized. Both bags started off fine but one has stalled and the myc does not appear to be growing. The second (somewhat) successful bag has healthy looking white mycellium growing throughout, I shake it and it all dissapears.

Now the questions:
1. How do I know when the 1 good bag is ready to spawn to hpoo?

2. How fast should the bags progress in relation to the jar?

3. What is the general opinion: Bags vs. Half gal. Jars?

4. Should I put off all attempts at cultivation until I have the equipment nessesary to do strain isolations, or is it okay to run with whatever the multispore LC gives me?

Side note: The control jar was a Non-Inoc'd control jar from a previous experiment to determine if I had a contaminated syringe. It was about 4 months old and has so far supported healthy and fast rizomorphic growth.

Edited : Bags are filled with WBS, soaked for 12 or so hours, when they quit expanding, and then drained of all excess moisture. No simmer.

1. you'll know because you wont see any grain, just white mycelium

2. too many variables, bags usualy take a little more than a month

3. I prefer the bags

4. no, Id simply review my procedures regarding the bag, list in this thread
anything you think that may have lead to the contams, it's apparently not the
syringe, did you tape the inoculation holes?

Thank you for your response;

Yes I taped the holes

I'm pretty sure I know why it contam'd....

Cause I don't have a flow hood.  I let the canner cool untill it gets to 2lbs, then relieve the rest of the preassure and get the lid off.
Quick as I can, I seal the bags by cinching the zip-ties that I put on the bags before I cooked them.

The first 2 over filled bags, one lost to wet spot one to greenies.
3rd bag I think is wetspot again...

BTW I have edited my original post to include that I am using WBS soaked and drained they way I read to do.

a couple things I see that can be wrong, try letting the PC cool with the bags in
them, dont use zip ties, outside air can enter through the small crevaces it
leaves any time you move the bag, if you can get a sealer, by all means do so. I
knocked up bags for a few years, never needed a flow hood to fo it.

Can't get a sealer right now, maybe in the future. What advice can be offered on sealing a mycobag without an impulse sealer?

When using the zip ties, I twist the open end and fold it back on itself, then almost completely close the zip tie. I'm pretty sure this is mostly airtight; the bags havn't burst and as soon as I get the ties cinched after cooking they promptly vaccuum themselves together and stay that way for a few days.

I usualy fold the top corners at 1/4, tape them to the bag, then fold the remainder of the top of the bag over a couple of times and tape it to the bag as well, of course I use a sealer now so I no longer have to use that method.

Ive folded the top over and covered it w/a sheet of paper and then used a hair straightener. A iron would probably also work. Just be careful to not leave the straightener/iron on one spot for too long, it'll melt the bag.
