Glove boxes are outdated and not common in modern fungal labs.
When designing a glove box, what is the significance of having the fann with a filter attached to it blowing into the glove box? > positive pressure in the glove box, comprised of "clean" air. what is the significance of having the fan with a filter attached to it blowing out of the glove box? >prevent back flow of air, however this is not absolutely necessary so long as you have some outlet for the "clean" air at an opposite end small enough to maintain positive pressure within the glovebox.
what is the significance of having no fan? >no airflow, this is only a good idea if you are not useing any flame and plan on fogging the glovebox with some sterilizing agent such as ethanol + some vircide (such as lysol).. and the fumes of that are not healty for your culture.. soo....
