Agar questions

Ok first, should i be asking agar questions in here or the advanced forum? (edit: well it was moved to advanced so thats that :P)
1: making agar: i have some MEA from fungi.com that says you should mix 50g to every 1 litre of water, then PC for 45min @ 15PSI. Despite the fact that just about every agar tek here says PC for 15-20min, i followed the 45min on the jar. Whats the deal with the massive variance on timing here?
2: condensation: everyone says a little is fine, but how much is too much? i am seeing almost 1cc of water sloshing around ontop of my agar
3: agar concentration: some people have said that they mix the agar with more water than directed and get faster growth.. what is the typical range on agar concentration that works? I also ask this because i made some dishes (actually jars because i am having contam issues with my multispore petris) and accidentally moved the decimal too many places and have 10% of the MEA powder as the label calls for. If for whatever reason this will work just fine i will use the shitload of jars i made, otherwise ill just make a new batch tonight.

dont toss it out, you can always use it for liquid cultures, some people say agars no good for lc but ive always used it. water condensation isnt really a problem as long as its clean, i pour my plates and the entire top gets covered in it. never caused a problem for me, the same with in the mix, ive had water on top of my agar in the bottle. not a problem as long as you keep it clean. just pour that out before you pour your plates. agar can be deluted pretty far and still be good. i accidently pressure cooked my agar too long and it wont solidify anymore, but ive got a tray of lions mane going right now on sloshy agar. so you're pretty much fine in my opinion. just do it right next time. good luck

1. depends on how messy you're gonna be. the steamed in the microwave has worked fine for me.
2. the less sloshing you do with the petri, the more condensation that will stay on the sides and lids of your jars/petri. big difference when you're talking 1cc and little spores and myc
3. don't know about water concentration, but I accidently pushed the top end of the peroxide range once and nothing would grow on it.

less sloshing? um, don't giggle it around when it's starting to colonize. think about it and watch what happens... The water droplets on the sides and top of the dish/jar trickle down to the agar. Leave it alone for a while and the water on the agar will evaporate and recondense on the lid and sides again away from the agar. Maybe I'm wrong, I've only started recently, but only a couple drops for your 1cc make a difference. Half and quarter pint jars work better in my opinion.

I have done agar in one of my jars which has a filter disc. I put it in my drying chamber to draw the water off. Other times I tip it towards the cap and unscrew upside down at an angle and shake the water out. In other jars I have sucked it out with a syringe.

hrmm. well my main issue is when doing multispore, plus i am having contam issues there so i have started up some agar jars to keep things more sterile. I could just throw some tyvek ontop with some breather holes and i bet that would fix alot

tyvek doesn't let water pass through very well, that's the point of it.
