Colonizing questions, for first time
    #4324021 - 06/22/05 10:41 AM (11 years, 5 months ago)

I just inoculated 20 cc of Treasure Coast spores the other day into 6 birdseed/verm 1/2 pints, and i was wondering about how long it takes before I might see anythign growing? I had to shuffle my jars around a bit because i changed up from a heating pad to a TiT system (which is maintaining a perfect 85 degrees i might add) , would exposing them to light for a brief while slow the process down or cause other problems? How prone to contamination would you say jars are during the handling process, moving them into new tubs? they only have needle holes in the tape still. thanks in advance

what ratios did you use for the cakes, did you use any specific tek? How are you preventing contams from getting in? Dry verm casing layer? Tyvek? need to know more. Germination should occur between 3 and 7 days though.

A little light won't effect the jars. Colonization depends on the strain and other variables. With that much spore solution, you may have added too much water to the substrate. That's a shitload of spores though, so if they do germinate, the jars should colonize fast. If you don't see anything after a full week, the syringe might have been a dud or there was a problem with your substrate mix.

Even if there isn't ANY sign of colonization? The environment is 78 degrees farenheit and there isnt any signs of water pooling nor is the substrate dryin up. I mean i can'tsee a single difference on the 5 of the 10 jars I have checked. These are Pink Buffalos from TheLittleGuy.com

average germination time is 3-5days... optimal incubation temp is 86, but above is 86 is worse than below. I always aim for between 84 and 86 degrees.

I am proventing contamination though a dry layer of verm on top, the tape is still left intact cept for needle holes and the tub has and is carefully cleaned with lysol. i may be seeing a little somthing happening, but its only day 2 1/2 and it may be my imagination. So higher than 86 is worse than low, good to know, ill keep a tight control on that. What can i do if i dont see any growth in the next week? Is there anyway to save the batch? thanks again

Day 6 multi/spore inoculate in optimal conditions.

Sometimes it takes longer than a week - to see signs of germination

No, don't bother mixing the bag. Just let it do its thing man, sometimes it takes a while. Especially if you have a whole bag of substrate to colonize. Just keep the conditions good and eventually you'll see some growth, even if it takes a week or so. I know it's hard waiting, but we don't kid when we say "patience is key".

Mix...

Do not panic until you get to about 2 weeks and "see no growth". Remember patience is a virtue! Dont expect a miracle until after you see pins

Dont incubate at 96% rH... thats just asking for failure right there. Optimal incubation is 70-80% rH and 86 degrees.
