Innoculating agar
    #432134 - 10/21/01 08:56 AM (23 years, 6 months ago)

Can one use a colonized rye grain to innoculate an agar dish?...my friend is wondering because his friend is always saying that there's left over grains stuck in the jar after they're cased. If not how come?

since agar can grow so many things on it and is so susceptible to contamination from outside sources, this is likely why many dont innoc. with grain to agar. it could work though, but you may have to do a lot of transfers to get a clean plate thats usable.

(HQ's curious, shes gonna try today and see. very unscientific, you know.)

If everythings sterile I don't see why it wouldn't work. But everything being sterile with only mush. mycelium would be the hard part (well to gauge anyhow.)

maybe im just really clean but even in open air agar innoculations from a variety of things, i do not have much contamination (i have had some contamination in innocculating the rye (yet this is in a glove box, go figure))
infact, i had left some agar a little too long and it had pinned majorly and grown some little shrooms, so i to just took a pair of tweezers , flamed it, grabbed the mini shroom and just took it out and popped it into a new dish, growing fine now. I even took an entire plate of agar (it had really dried, but the agar was still holding it together) and popped into a dish, again in open air. doing nicely....

maybe im just lucky withthe agar and the rye hates me...
