tryptamine issues

Tryptamine hydrochloride is highly insoluble in water, even with heating. It is, however, readily solube in DMSO. However, even according to existing threads, faqs and primary literature, it has been experienced on numerous attempts that colonization does not occur, occurs slowly, or contaminates while those inoculated without the tryptamine colonize perfectly and fruit. Since the tryptamine is a crystal, it seems unlikely to be effective as an additive not equally distributed by solution in the substrate. Does anyone have any ideas or suggestions or experiences to help?

Yes! Tryptamine is an inhibitor (of the tryptophan decarboxylase enzyme not of "psilo production" as you name it).

The specification sheet for tryptamine hydrochloride from Sigma-Aldrich states that the solubility in water is 1% (although not at which temperature - I'm guessing 25 C). Gartz used 25 mM in his experiment, which corresponds to about 0.5%. I do not see a problem here. Even if the tryptamine had been in the solid state, it could still have been mixed well into the substrate, preferably as fine particles and not as one big crystal though.  :smirk:

I thought about powdering it myself, although wondered about the resultant distribution throughout the mycelial network. Gartz's 25mM, of course, represents a solution which is not easily obtained since the material is largely insoluble - i.e. take 25mg and dissolve in 1liter of waterand you have a 25mM solution, but not if 99% is undissolved. See the quandary?

I think you might have misunderstood the solubility. A 1% solubility does not mean that 99% of the substance will stay undissolved regardless of the amount of solvent, but rather that 1 part of the substance will dissolve in 99 parts of the solvent. So in 1 liter (or to be nit-picky: 990 grams) of water, one can dissolve approximately 10 grams of tryptamine hydrochloride.

You might want to check up on molarity before you start mixing. Your calculation is not correct.
