Colonization problems???

When incubating jars the same process is used for preparing WBS jars. Soaked for 12-36 hrs. simmered until signs of cracked seeds are present. Then drained until no water droplets are dripping from strainer then pc'ed for 1 hour left to cool, then inoculated with 2cc's. then incubated at 86 degrees. And maybe 15% of the time a successful jar is the outcome. What is the deal??? Could it be weak spores or something this very frustrating since I have every other step down, but unsuccessful jar attempts is holding this project back.

Are you using direct spores or a liquid culture?

What are you using as for gas exchange - in the jar lids.

85% failure is related to something.

Thats a difficult one. I have never used wbs before, but I once read something about a fungicide or something that certain companies put on their seed. It will inhibit the growth of mycelium, thus could explain the problem that you are having. I have used cracked corn before with excellent resluts and its cheaper then wbs, if all else fails maybe you could try using that.

I have 2 holes in the lid but the lid is covered with postal tyvek, then after metal ring is inplace I put a coffee filter over it.

I am basically in the same situation. So far I've lost 2-3 jars to contams and the other 6-7 are showing little to no signs of colonization.

It'd be nice to know that you need holes for air exchange BEFORE reading teks. Why didn't they put that in there?
