LC jar lids...attaching tubing sections with RTV HT stuff...help!

    So I've tried to attach latex tubing (bungee cord surgical type stuff) with the RTV high-temp gasket maker stuff.  I'm having some problems, though, as the gasket goo doesn't seem to bond well with the latex tubing or the jar lid.  Just tried to do some jars of LC up and the goo came out looking like it wasn't sealed still. 
    Questions, then:
    I have a 7 quart PC that just fits quart jars; with tubing on top this PC lid presses down and bends the tubing while it cooks.  Is this stress causing the problem? 
    Is there a way to make the bond stronger between these materials and the gasket maker goo? 
    Is there a better type of tubing (e.g. catheter tubing) that would be better as its diameter is smaller and thus is less resistant to the PC lid bending and also is easier to fix in with the gasket maker because it's smaller than surgical tubing? 

    This is sort of for Agar, but, anyone who can help, please do.  :laugh:

You guys make simple things ultra complicated, and it kills me. (not you personally) All you need for a good liquid culture lid is a 1/4" drilled hole stuffed with polyfill, and another hole big enough for a 16-20 gauge needle. Insert 1-2cc spores, cover with tape. Once colonized, wipe tape with alcohol and draw liquid culture right into sterilized syringe.

It's the swishing that's a problem...I don't want to have a filter that can get soaked.  Maybe I'll just have to be careful about that and use some tyvek as it appears I'M A FUCKING RETARD with tubing.  :smirk:

Use a shouldered jar, and a gentle swirl shouldn't cause you any problems. I never shake a LC, just a swirl once or twice during the cycle is all you need.

It may be the LATEX TUBING... is the problem.

I use rubber tubing (urine cathiter type) & have never had a problem with RTV gasket material sticking to the tubibg, or lid.

i used Agar's advice and used windshield washer wiper tubing and didn't have any problems,



using the same sealant you are.

is this the best way to start off pan cyan?... to start a liquid culture with spore syringe first?.. or is it just as good to go ahead and squirt the spores on WBS?... and what liquid media recipe is best for pan cyan/??

I think I got a few feet of it for $1.59. Should be able to get it at any auto part store. I got mine at AutoZone.

Not sure what is best for pan cyan/, if it would require anything special, but I like to use a tablespoon each of light malt extract and dextrose, $6 for both and enough to last a lifetime,



per quart of distilled water. PC'ed for 20 minutes at 15 psi.

I got that same stuff.  Must say...best LC I've seen from that combo.  One tablespoon each to a full quart of warm water, mix, pc 20 minutes, strain through a coffee filter, fill quart jars to about 1/2 ~ 3/5 full, pc 20 again.  Very nice myc. in a week or two.  :sun:  :thumbup: Cheers agar.  :laugh:
