in this link it says spores will germinate on antibiotic agar, is this true for all antibiotics, i have amp and kan not sure of the full names, will these work for germinating azure or ps.cyanofibrillosa spores?

I was under the impression that you transferred to antibiotic agar once you have a good sample of mycelium, as spores will not germinate on antibiotic agar.

I haven't yet experimented with agar, however, so i couldn't be 100% sure.

Looks like I found your answer in your own post...and it confirms my earlier reply:

Many people seem a bit confused as to what peroxide can and can not do. Hydrogen peroxide will kill bacteria and fungal spores and inhibit the growth of yeasts. ----Unfortunately, mushroom spores fall under the category of fungal spores, so they will not germinate on peroxidated agar because they are killed on contact.---- The good part about this is that mold spores, such as spores from the dreaded green mold, fall under this category as well and are also killed upon contact with the agar surface.

Actually thats not the answer that he was looking for.

I can understand your wanting the topic moved to advanced after a few replies that didnt help at all (sorry guys, but if you dont know what your responding to, you should respond), but be a little more patient, its only been up an hour or so, many havnt been on to look here. There are many people who know much on the subject who come to these forums, you just have to give it a little more time. Again though, I understand the frustration when you just get replies from people who havnt used agar or "buy some".

You can germinate cubies on kanamycin but I'm unsure of the other species. They should be ok, but I dont know if you'll find someone whos done that species with that antibiotic.

AMP is antimicrobial peptides, and there are many kinds...do you know what kind yours is? Some are antifungul so probably wouldnt work. If antibacterial then it should also allow spores to germinate. As I said though, exotic species of mycelium is a bit different and can be tough to germinate on antibiotic agar...I would try both, and some with out..

Peroxide kills spores, but not all antibiotics do.

I germinate the spores of all kinds of mushrooms species on antibiotic agar, including P. azurescens, P. cyanescens, and P. cyanofibrillosa. I most often use gentamycin sulfate.

Hydrogen peroxide, although it inhibits the growth of many microorganisms, is not an 'antibiotic' in the strict sense of the word.

However still that part of his post wasnt really needed (which is why I missed it, skimmed over it)...the important part is the antibiotics which he has and if they'll work..

yah in the link i posted it DID say spores will germ on antibiotics but like so many ppl posting here i was un sure, i will try the kan with a few dishes and see if they will help clean up these wild prints i have...this should be in the archives..

heres what i was refering to...

A good idea would be to order some antibiotic agar from FP to use for germinating spores. This stuff, while resistant to bacteria, is still susceptible to contamination by mold and yeast, so don't forget to use the proper sterile techniques (like a glove box, bleached down room, or a warm oven). Once the spores germinate, the mycelium can be transferred to peroxidated agar with less of a stress on sterility. It is important to remember to keep your scalpels clean and to thoroughly clean and sterilize your plates prior to pouring! If there are small mold colonies left in the plates when you pour them, they will be resistant to the peroxide and will grow.

ya i just read that, my apologies. I thought the whole reason they sold antibiotic AND non-antibiotic is because u couldnt germinate on antibiotic, because then why would anyone even use non-antibiotic...But i think i may be associating "peroxided" and "antibiotic" too closely.

which leads me to my next conclusion, antibiotics can be used to help clean up dirty prints a holelot better then regular agar....please archive this...
